Dimethyl Sulfoxide is a dipola, aprotic solvent. It has been shown to accelerate strand renaturation (1-10% concentration) and is believed to give the nucleic acid thermal stability against depurination. Dimethyl Sulfoxide is used to enhance dermal absorption of many chemicals, as a solvent for many organic and inorganic compounds including fats, carbohydrates, dyes, resins, and polymers, in antifreeze or hydraulic fluids, as a cryopreservative for cell cultures, oxidation of thiols and disulfides to sulfonic acids, as a PCR cosolvent to help improve yields, especially in long PCR. DMSO is routinely used in polymerase chan reaction (PCR), amplification of cDNA libraries, DNA sequencing, column-loading buffers for poly (A)+ RNA selection, buffers for the transformation of competent E. coli, and transfection protocols. To prepare sterile solutions use a teflon or nylon membrane to sterile-filter the DMSO - do not use a cellulose acetate membrane.

CD152 and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

It recognises a polypeptide of 6.5 kDa, identified as pS2 estrogen-regulated protein. Its epitope is localized between aa57-84 of human pS2 protein. pS2 is a trefoil peptide. Trefoil peptides are protease resistant molecules secreted throughout the gut that play a role in mucosal healing. These peptides contain three intra-chain disulfide bonds, forming the trefoil motif, or P-domain. pS2 is known to form dimers and this dimerization is thought to play a role in its protective and healing properties. About 60% of breast carcinomas are positive for pS2. Staining is cytoplasmic, often with localization to the Golgi apparatus. pS2 is shown to be localized in normal stomach mucosa, gastric fluid, goblet cells in the colon and small intestine, and in ulcerations of the gastrointestinal tract. Several studies have shown that pS2 is primarily expressed in estrogen receptor-positive breast tumors and it may define a subset of estrogen-dependent tumors that displays an increased likelihood of response to endocrine therapy.

β-Galactosidase assay is for determining the β-galactosidase activity in the lysates of cells transfected with a β-galactosidase expression construct. It uses chlorophenol red-β-D-galactopyranoside (CPRG), which is up to 10 times more sensitive than the classic O-nitrophenyl-β-D-galactopyranoside (ONPG) β-Galactosidase assays. This increased sensitivity makes it simpler to measure β-galactosidase activity in cells that are difficult to transfect or have low β-galactosidase activity expression. β-Galactosidase activity is measured using a spectrophotometer or a microplate reader to determine the amount of substrate converted at 570 to 595 nm.
