The UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase (GalNAc-T) family of enzymes are substrate-specific proteins that catalyze the transfer of GalNAc (N-acetylgalactosamine) to serine and threonine residues onto various proteins, thereby initiating mucin-type O-linked glycosylation in the Golgi apparatus. GalNAc-T13 (Polypeptide N-acetylgalactosaminyltransferase 13), also known as UDP-GalNAc:polypeptide N-acetylgalactosaminyltransferase 13, is a 556 amino acid protein that displays much stronger enzymatic activity than GalNAc-1 towards GalNAc transfer to mucin peptides such as Muc5a and Muc7. The N-terminal domain is involved in substrate binding and manganese coordination, while the C-terminal domain is involved in UDP-Gal binding and catalytic reaction. With specific expression in the central nervous system, GalNAc-T13 may be responsible for the synthesis of Tn antigen in neuronal cells, which is a universal carcinoma marker on malignant cells.

Sulfation is an essential conjugation reaction that increases the water solubility of many compounds, thereby influencing their renal excretion and also resulting in the formation of active metabolites. SULT4A1 (sulfotransferase family 4A, member 1), whose alternative names include brain sulfotransferase-like protein, nervous system sulfotransferase, NST, SULTX3, hBR-STL-1, BRSTL1, BR-STL-1, MGC40032 and DJ388M5.3, is a 284 amino acid protein showing cytoplasmic localization. As a member of the sulfotransferase 1 family, SULT4A1 plays a role in elimination of xenobiotics, activation of procarcinogens and regulation of hormones. SULT4A1 is highly expressed in cerebral cortex and frontal lobe, with lower expression in cerebellum, temporal and occipital lobes. Two SULT4A1 isoforms exist to alternative splicing events. The gene encoding SULT4A1 maps to human chromosome 22q13.2, a region which has been implicated in predisposition to schizophrenia.

The Hox homeobox genes encode proteins that are transcriptional regulators with an established role in embryonic development. HoxA4 (homeobox A4), also known as HOX1D or HOX1, is a 320 amino acid protein that localizes to the nucleus and contains one homeobox DNA-binding domain. Expressed in the embryonic nervous system, HoxA4 functions as a sequence-specific DNA-binding transcription factor that is part of a regulatory mechanism that provides cells with positional identities during development. Via its ability to bind DNA, HoxA4 plays an important role in the regulation of gene expression, as well as morphogenesis and differentiation. The gene encoding HoxA4 maps to human chromosome 7, which houses over 1,000 genes and comprises nearly 5% of the human genome. Defects in some of the genes localized to chromosome 7 have been linked to Osteogenesis imperfecta, Williams-Beuren syndrome, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome.

Probably acts as an adapter that recognizes polyubiquitinated misfolded proteins and target them to the aggresome, facilitating their clearance by autophagy.

The Eph subfamily represents the largest group of receptor protein tyrosine kinases identified to date (1–3). While the biological activities of these receptors have yet to be determined, there is increasing evidence that they are involved in central nervous system function and in development (1–3). The Eph subfamily receptors of human origin (and their murine/avian homologs) include EphA1 (Eph), EphA2 (Eck), EphA3 (Hek4), EphA4 (Hek8), EphA5 (Hek7), EphA6 (Hek12), EphA7 (Hek11/MDK1), EphA8 (Hek3), EphB1 (Hek6), EphB2 (Hek5), EphB3 (Cek10, Hek2), EphB4 (Htk), EphB5 (Hek9) and EphB6 (Mep). Ligands for Eph receptors include ephrin-A4 (LERK-4) which binds EphA3 and EphB1. GPRC6A (G protein-coupled receptor family C group 6 member A) is a 928 amino acid protein encoded by the mouse Gprc6a gene. GPRC6A is a member of the G protein-coupled receptor 3 family. This receptor is activated by both amino acids and extracellular concentration of calcium ions. The activity of GPRC6A is mediated by a G protein that activates a phosphatidylinositol-calcium second messenger system. GPRC6A senses changes in the extracellular concentration of calcium ions, suggesting that it may mediate extracellular calcium-sensing responses in osteoblasts. Osteocalin stimulates the activity of GPRC6A in the presence of calcium. GPRC6A also acts as a receptor for amino acids, with a preference for basic amino acids such as L-Lys, L-Arg and L-ornithine. Its affinity for amino acids suggests that it may act as a regulatory component of the urea cycle. GPRC6A is expressed at high levels in liver, lung, spleen and heart, and at lower levels in kidney, skeletal muscle and brain.

ApoD is mainly associated with high density lipoproteins in human plasma. ApoD is expressed in numerous tissues having high levels of expression in spleen, testes and brain. ApoD plays a role in maintenance and repair within the central and peripheral nervous systems. ApoD occurs in the macromolecular complex with lecithin-cholesterol acyltransferase. It is a multi-ligand, multi-functional transporter and transports a ligand from 1 cell to another. ApoD is probably involved in the transport and binding of bilin, it appears to be able to transport a variety of ligands in a number of different contexts.

SPRYD5 Antibody: The SPRY domain-containing protein 5 (SPRYD5) is a member of a family of proteins whose sole common characteristic is the presence of a SPRY domain. SPRY domains are structural domains that were first described in the fungal Dictyostelium discoideum tyrosine kinase spore lysis A. In most systems SPRY domains provide binding sites for regulatory proteins or intramolecular binding sites that maintain the structural integrity of a protein. SPRYD5 belongs to the TRIM/RBCC family and contains one B box-type zinc finger, one B30.2/SPRY domain and one RING-type zinc finger. Little is known of the function of the SPRYD5 protein.

CD5 Antigen-Like (CD5L) is a soluble protein that belongs to group B of the scavenger receptor cysteine-rich (SRCR) superfamily and contains three SRCR domains. CD5L is a secreted glycoprotein and expressed by macrophages presentin lymphoid tissues. It binds to myelomonocytic and lymphoid cells and may play an important role in the regulation of the innate and adaptive immune systems. CD5L functions as a pattern recognition molecule by binding both lipoteichoic acid (LTA) on Gram positive and lipopolysaccharide (LPS) on Gram-negative bacteria and the SRCR domain one of CD5L retains both the LPS and LTA binding activities. Furthermore, CD5L seems to play a role as an inhibitor of apoptosis.

Testican-2 is unique in that it actually abolishes the inhibition of MT-MMPs by other testican family members and specifically inactivates N-Tes by binding to its COOH-terminal extracellular calcium-binding domain. Testican-2 halts neurite growth from cerebellar neurons and may be involved in regu-lating the development of the CNS.

HLA-A belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exons 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region, and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Calcipressin-2, also known as RCAN2 and DSCR1L1 (Down syndrome candidate region 1-like 1), is a 197 amino acid protein that is highly expressed in both the cytoplasm and nucleus of neurons and is the only RCAN family member that is found in glial cells. Calcipressin-3, also known as RCAN3 and DSCR1L2 (Down syndrome candidate region 1-like protein 2), is a 241 amino acid protein that potentially is involved in central nervous system development. Calcipressin-3 interacts with cardiac troponin I, suggesting that it may play a role in cardiac contraction events.
