The insulin receptor related receptor (IRR) is a heterotetrameric transmembrane receptor composed of two alpha and 2 beta chains linked by disulfide bonds. The alpha chains contribute to the formation of the ligand-binding domain, while the beta chains carry the kinase domain. Member of the insulin RTK family, IRR shares high homology with the insulin (IR) and the insulin-like growth factor-1 receptor (IGF-1R), but doesn’t bind any of IR and IGF-1R known ligands. In contrast to the widespread patterns of expression to IR and IGF-1R, IRR demonstrates a very restricted cellular distribution in a subset of tissues of neuronal origin and its biological functions are still unknown.

The protein encoded by this gene catalyzes the formation of inositol 1,4,5-trisphosphate and diacylglycerol from phosphatidylinositol 4,5-bisphosphate. This reaction uses calcium as a cofactor and plays an important role in the intracellular transduction of receptor-mediated tyrosine kinase activators. For example, when activated by SRC, the encoded protein causes the Ras guanine nucleotide exchange factor RasGRP1 to translocate to the Golgi, where it activates Ras. Also, this protein has been shown to be a major substrate for heparin-binding growth factor 1 (acidic fibroblast growth factor)-activated tyrosine kinase. The membrane-bound isoform 1 is a receptor involved in the development of the cardiovascular system, in angiogenesis, in the formation of certain neuronal circuits and in organogenesis outside the nervous system. It mediates the chemorepulsant activity of semaphorins. It binds to semaphorin 3A, The PLGF-2 isoform of PGF, The VEGF-165 isoform of VEGF and VEGF-B. Coexpression with KDR results in increased VEGF-165 binding to KDR as well as increased chemotaxis. It may regulate VEGF-induced angiogenesis. The soluble isoform 2 binds VEGF-165 and appears to inhibit its binding to cells. It may also induce apoptosis by sequestering VEGF-165. May bind as well various members of the semaphorin family. Its expression has an averse effect on blood vessel number and integrity.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). Long pentraxins are a family of highly conserved proteins that are expressed in the brain and central nervous system, and form multimeric complexes. Neuronal pentraxin 1 (NP1), NP2, and neuronal pentraxin receptor (NPR) are members of the long pentraxins that represent a neuronal uptake pathway that may function during synapse formation and remodeling. The NP1 gene is located on chromosome 17q25.3 and the protein product mediates the uptake of synaptic material, including the presynaptic snake venom toxin, taipoxin (3). NP2, whose function is unknown, is located on chromosome 7q22.1 and like NP1 contains several potential N-linked glycosylation sites. TPPP (Tubulin polymerization-promoting protein), also known as p24 and p25, is a widely expressed 219 amino acid protein found in the perinuclear region of the cytoplasm. TPPP may form dimers and functions in polymerizing tubulin into double-walled tubules, polymorphic aggregates, or stabilized blocks. TPPP overexpression prevents formation of the mitotic spindle assembly and breakdown of the nuclear envelope. The 3-HAO family of proteins consists of intramolecular dioxygenases that contain non-heme ferrous iron. HAAO (3-hydroxyanthranilate 3,4-dioxygenase), also known as HAO or 3-HAO, is a 286 amino acid monomeric cytoplasmic protein that belongs to the 3-HAO family. Utilizing Fe²⁺ ions as a cofactor, HAAO participates in the formation of quinolinic acid (QUIN), and excitotoxin, by catalyzing the oxidative ring opening of 3-hydroxyanthranilate to 2-amino-3-carboxymuconate semialdehyde. HAAO may play a role in disorders that are associated with varying levels of QUIN expression, and is widely expressed in peripheral organs, including kidney and liver. Existing as two alternatively spliced isoforms, the gene encoding HAAO maps to human chromosome 2p21 and mouse chromosome 17 E4.
