Medium 199 wurde entwickelt um den Nährstoffbedarf von embryonalen Hühnerfibroblasten zu untersuchen. Heute wird Medium 199 häufig für die Erhaltung nicht transformierter Zellen, die Impfstoff- und Virusproduktion und primäre Explantate von Epithelzellen verwendet. Die mit Hanks-Salzen formulierten Medien sind mit Salzlösungen gepuffert und für den Ausgleich bei Umgebungsbedingungen ausgelegt. Ihre Verwendung in einem CO₂-Inkubator führt zu einem schnellen Abfall des pH-Werts des Kulturmediums.

McCoy’s media were originally formulated for growth and support of lymphocytes. This final modification produced a medium identical to RPMI 1629. McCoy's 5A media supports the indefinite proliferation of Walker 256 carcinoma cells. In addition, it is excellent for the propagation of leukocytes, biopsy tissues, a broad range of human and rat normal or transformed cell types, the most current primary and continuous cell lines.

MEM is an advancement of the BME and the base medium of many further modifications. Because BME did not fulfil all requirements for some mammalian and HeLa cells, a better variation had to be developed. Today, MEM is one of the most used synthetic media and shows its versatility by supplementing with amino acids including Hank's or Earle's salts. Even the addition of only small amounts of FBS results in a positive effect on cell growth.

In 1976, Guilbert and Iscove demonstrated that precursor cells of erythrocytes and macrophages could be cultured in a reduced-serum medium supplemented with albumin, transferrin, lecithin, and selenium. Iscove's medium is a modification of Dulbecco's Modified Eagle's Medium (DME) containing selenium, additional amino acids and vitamins, sodium pyruvate, HEPES buffer, and potassium nitrate instead of ferric nitrate. Further studies have demonstrated that Iscove's medium will support murine B lymphocytes, haemopoietic tissue from bone marrow, B-cells stimulated with lipopolysaccharide, T-lymphocytes, and a variety of hybrid cells.
