Microtubules mediate the spatial organization of diverse membrane-trafficking systems. The HOOK proteins, HOOK1, HOOK2 and HOOK3, comprise a family of cytosolic coiled-coil proteins that contain conserved N-terminal domains, which attach to microtubules; and more divergent C-terminal domains, which mediate binding to organelles. HOOK1, a cytoskeletal linker protein, may play a role in endocytic membrane trafficking. It exists as a homodimer, most likely mediated through its central coiled-coil domain. HOOK1 interacts with VPS18 and is required for spermatid differentiation, in which it is most likely involved in the positioning of the manchette microtubules and the flagellum. HOOK1 localizes primarily to the cytoplasm and does not associate with the Golgi complex, unlike HOOK3, which participates in the organization of the cis-Golgi compartment.

The stain uses an enhanced Periodic Acid-Schiff (PAS) method for detection of glycoprotein sugars. The supplied agent first oxidises the cis-diol sugar groups to aldehydes. The aldehyde groups react with the sensitive glycoprotein stain solution forming Schiff bonds and producing strong magenta colour bands. RAPIDstain™ can be used after glycoprotein staining to detect non-glycosylated proteins and the use of the stain enhances glycoprotein staining. The retinal pigment epithelium (RPE) is a monolayer simple epithelium in proximity to the outer surface of the retinal photoreceptor cells. Retinal pigment epithelium-specific protein (RPE65) is a 65 kDa protein belonging to the β-carotene dioxygenase family. This protein is important in 11-cis retinal production as well as in visual pigment regeneration. RPE65 is attached to the membrane by a lipid anchor when palmitoylated (membrane form) and soluble when unpalmitoylated. The soluble form of the protein binds vitamin A. Defects in RPE65 causes autosomal dominant retinitis pigmentosa and/or Leber congenital amaurosis type 2.

Pin1 is a Peptidyl-prolyl isomerases (PPIase). Peptidyl-prolyl isomerases (PPIase) facilitate the cis-trans interconversion of the peptidyl-prolyl bond thereby affecting protein folding. Pin1 is a PPIase which specifically recognizes phosphorylated S/T-P bonds. Pin1 has been implicated in tau pathologies that underlie Alzheimer's Disease. Pin1 binds to tau phosphorylated specifically on the Thr231-Pro site and induces conformational changes in tau. Such conformational changes can directly restore the ability of phosphorylated Tau to bind microtubules and promote microtubule assembly and/or facilitate tau dephosphorylation. Pin1 expression inversely correlates with the predicted neuronal vulnerability in normally aged brain and also with actual neurofibrillary degeneration in AD brain. Pin1 could be pivotal for maintainance of normal neuronal function and preventing age-dependent neurodegeneration.
