The Annexins constitute a family of structurally-related, relatively abundant proteins that exhibit Ca2+-dependent binding to phospholipids. Annexins function in multiple aspects of cell biology including regulation of membrane trafficking, transmembrane channel activity, inhibition of phospholipase A2, inhibition of coagulation and mediation of cell-matrix interactions. Annexin A13 is considered the original progenitor of the 12 members of vertebrate Annexins. The expression of Annexin A13 is highly tissue-specific, being expressed only in intestinal and kidney epithelial cells. This expression is associated with a highly differentiated intracellular transport function. Two alternative splicing isoforms of Annexin A13 exist, both of which bind to rafts.

Existing as a homodimer, PHC1 contains one FCS-type zinc finger and a SAM (sterile alpha motif) domain. PHC1 is encoded by a gene located on human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome.

Membrane fusion requires interaction among SNARE1 proteins associated with both donor and acceptor membranes (1,2). Following membrane fusion, the α-SNAP cytoplasmic adapter protein binds to the SNARE complex. N-ethylmaleimide-sensitive factor (NSF), a hexameric ATPase, then associates with the α-SNAP/SNARE complex to mediate SNARE disassembly during membrane fusion (3,4). The ATPase activity of NSF induces a conformational change in the α-SNAP/SNARE complex that leads to its dissociation from the membrane, membrane fusion and eventual recycling of the SNARE complex for subsequent membrane fusion (3,4).

eIF3K (Eukaryotic translation initiation factor 3 subunit K, Muscle-specific gene M9 protein) is a widely expressed translation initiation factor that belongs to the eIF3 subunit K family. Translation initiation factor 3 (eIF3) is a multisubunit complex containing at least 12 subunits. eIF3 binds to the 40S ribosomal subunit, promotes the binding of methionyl-tRNAi and mRNA, and interacts with several other initiation factors to form the 40S initiation complex. eIF3K is the smallest subunit of eIF3 and it interacts with several other subunits of eIF3 and the 40S ribosomal subunit. eIF3K is conserved among high eukaryotes, including mammals, insects, and plants, and it is ubiquitously expressed in human tissues. eIF3K is distributed both in nucleus and cytoplasm and colocalizes with cyclin D3, a regulatory subunit of cyclin-dependent kinase 4 (Cdk4).

Lymphotactin is a chemokine-like molecule produced selectively, if not exclusively, by activated CD8+ T cells (1). Lymphotactin is a C-type member of the chemokine superfamily, but retains only the 2nd and 4th of the four cysteine residues conserved in other chemokines (3,4). In normal tissues, lymphotactin is expressed at high levels in spleen, thymus, small intestine, and peripheral blood leukocytes, as well as at low levels in lung, prostate, and ovary (2). Lymphotactin is chemotactic for CD4+ and CD8+ T cells but not for monocytes, and induces a rise in intracellular calcium in peripheral blood lymphocytes (1). The specific receptor for lymphotactin is XCR1, a member of the G protein-coupled receptor family (5). The gene which encodes lymphotactin maps to human chromosome 1q23 (1,2).

Localized to the nucleus, GIOT-1 is widely expressed in tissues, with highest levels in liver, kidney, small intestine, pancreas and thymus. GIOT-1 contains 12 C2H2-type zinc fincers and one KRAB domain. Because the KRAB domain functions as a transcriptional repressor when attached to the template DNA, GIOT-1 is thought to be involved in transcriptional regulation. The gene encoding GIOT-1 is localized to chromosome 19q13.12 and two isoforms of GIOT-1 exist as a result of alternative splicing events.

FGFR1OP2 belongs to the SIKE family. The FGFR1OP2 (FGFR1 oncogene partner 2) gene was identified through its involvement in a fusion with the FGFR1 gene. FGFR1OP2 may be involved in the wound healing pathway. It is expressed in bone marrow, spleen and thymus. A chromosomal aberration involving FGFR1OP2 may be a cause of stem cell myeloproliferative disorder (MPD). Insertion ins(12;8)(p11;p11p22) with FGFR1. MPD is characterized by myeloid hyperplasia, eosinophilia and T cell or B cell lymphoblastic lymphoma. In general it progresses to acute myeloid leukemia. The fusion protein FGFR1OP2-FGFR1 may exhibit constitutive kinase activity and be responsible for the transforming activity.

Das B2m ELISA-Kit (Maus) (OKEH00344) von Aviva Systems Biology basiert auf der standardmäßigen Sandwich-Enzym-Linked-Immunosorbent-Assay-Technologie. Ein für B2m spezifischer Antikörper wurde auf eine 96-Well-Platte (12×8-Well-Streifen) vorbeschichtet. Standards oder Testproben werden in die Vertiefungen gegeben, inkubiert und entnommen. Ein für B2m spezifischer biotinylierter Detektorantikörper wird hinzugefügt, inkubiert und anschließend gewaschen. Anschließend wird das Avidin-Peroxidase-Konjugat zugegeben, inkubiert und nicht gebundenes Konjugat abgewaschen. Durch die Zugabe von TMB-Substrat wird eine enzymatische Reaktion erzeugt, die durch HRP katalysiert wird und ein blaues Farbprodukt erzeugt, das sich nach Zugabe einer sauren Stopplösung in Gelb ändert. Die anhand der Absorption bei 450 nm abgelesene Dichte der Gelbfärbung ist quantitativ proportional zur Menge der in der Vertiefung eingefangenen Probe B2m.

Neuroligins are a family of plasma membrane proteins that possess an N-terminal hydrophobic domain, a large esterase homology domain, a single transmembrane region, a short cytoplasmic domain, and an EF-hand binding domain (1,2). Members of the neuroligin family include Neuroligin 1, Neuroligin 2 and Neuroligin 3. Neuroligins are expressed in excitatory neuronal synaptic clefts. Neuroligins play a role in the formation and remodeling of CNS synapses by binding to b-neurexins, a family of neuronal cell surface proteins. Neuroexin 1b binds to the EF-hand domain of Neuroligin 1 and requires calcium ion. Neuroligins also bind to PSD-95, which may recruit ion channels and neurotransmitter receptors to the synapses.
