Beschreibung:   Retinoic acid (RA; active metabolite of vitamin A) plays a prominent role in regulating the transition of proliferating precursor cells (such as carcinoma cells and neuronal precursors) to postmitotic differentiated cells (Joshi et al., 2005). The retinoid X receptors (RXRs) family (RXRalpha, beta and gamma), preferentially bind 9-cis-RA and regulate gene transcription by forming heterodimers with a second family of RA receptors. RAs have been suggested to potentially play a therapeutic role in cervical cancer (Abu et al., 2005). RAs are known to play key roles in neuronal development and an increasing body of evidence indicates that retinoid signaling may regulate synaptic plasticity and associated learning and memory behaviors (Lane and Bailey, 2005).

Beschreibung:   Ferritin heavy polypeptide 1(FTH1), is a ubiquitous intracellular protein which stores iron in a soluble, non-toxic, readily available form. FTH1 has ferroxidase activity and is important for iron homeostasis. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Ferritin is composed of 24 subunits of the light and heavy ferritin chains. It plays a role in delivery of iron to cells and mediates iron uptake in capsule cells of the developing kidney. Variation of ferritin subunit composition may affect iron absorption and release in different tissues. Deficiency of ferritin proteins may cause several neurodegenerative diseases. Almost all living organisms can produce this protein, including algae, bacteria, higher plants, and animals.

Beschreibung:   Following restriction enzyme digestion of DNA molecules, researchers need to rejoin the ends of the DNA to generate recombinant DNA, a process known as ligation. Ligation of DNA is achieved with the bacterial enzyme T4 DNA ligase, which catalyses the formation of phosphodiester bonds. The DNA Ligation kit teaches students about ligation as they ligate several DNA fragments together to make larger pieces of DNA that are easily identified by agarose electrophoresis.

Beschreibung:   Dialysis is a routinely used technique in research laboratories to “change” the solution a biomolecule is dissolved in. Often the buffers used to isolate biomolecules, such as proteins, are not compatible with downstream applications due to high concentrations of reagents, such as salts. A sample is placed in a bag with a semi-permeable membrane and then placed in the new solution or water. Small molecules, such as salt, can pass through the membrane, moving from an area of high concentration to low concentration. The larger molecules, such as proteins, are retained in the bag. Microdialysis is a modification of dialysis to overcome the problem of dialysing small volumes of precious samples. Microdialysis uses small devices designed for small volumes. This kit allows students to study dialysis using patented microdialysis devices.

Beschreibung:   CLEC7A Antibody: CLEC7A, also known as dectin-1, is a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily and is predominantly expressed on myeloid cells. It is a small glycoprotein type II membrane receptor with an extracellular C-type lectin-like domain fold and a cytoplasmic domain with an immunoreceptor tyrosine-based activation motif (ITAM). CLEC7A functions as a pattern-recognition receptor that recognizes a variety of beta-1,3-linked and beta-1,6-linked glucans from fungi and plants, and in this way plays a role in innate immune response. Upon fungal exposure, CLEC7A activates Syk tyrosine kinase, triggering a massive oxidative burst through the formation of reactive oxygen species.

Beschreibung:   The Gram staining method was first described in 1844 by the Danish bacteriologist Hans Christian Gram, after whom the test was named. The Gram staining test for bacteria is one of the most important tests in microbiology and is often one of the first tests performed in the identification of bacteria. The Gram staining method utilises the properties of bacterial cell walls and the stain crystal violet. Gram-positive microorganisms have a higher peptidoglycan and lower lipid content than Gram-negative bacteria in their cell walls and this makes them capable of retaining the Gram stain. This kit is supplied with two strains of bacteria and all the necessary components to carry out the Gram staining.

Beschreibung:   Cefotaxime is a cephalosporin, which acts by interference of synthesis of peptidoglycan of the bacterial cell wall. Cefotaxime, a cephalosporin antibiotic, is active against both gram-negative and gram-positive bacteria. In addition, cefotaxime blocks the division of cyanobacteria and lower plant organelles/plastids such as the photosynthetic organelles of Glaucophytes and chloroplasts of bryophytes.
Cefotaxime is used for infections of the respiratory tract, skin, bones, joints, urogenital system, meninges, and bloodstream. It generally has good coverage against most Gram-negative bacteria, with the notable exception of Pseudomonas. It is also effective against most Gram-positive cocci except for Enterococcus. It is active against penicillin-resistant strains of Streptococcus pneumoniae. It has modest activity against the anaerobic Bacteroides fragilis. In meningitis, cefotaxime crosses the blood–brain barrier better than cefuroxime.
Cefotaxim inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs) which inhibits the final transpeptidation step of peptidoglycan synthesis in bacterial cell walls. As a result, bacteria lyse due to cell wall autolytic enzymes.

Beschreibung:   PNAS4 Antibody: PNAS4 is a highly conserved protein that shares high homology from plants to animals. It was initially identified as a putative apoptosis-related protein in the human acute promyelocytic leukemia cell line NB4. PNAS4 is activated during the early response to DNA damage and can inhibit proliferation via apoptosis when overexpressed in some tumor cells such as U2OS, SKOV3, and A549. PNAS4 inhibits tumor cell proliferation through the following mechanisms: (i) overexpression of PNAS4 causes S phase arrest by regulating the expression of cell cycle-related proteins and (ii) PNAS4 induces apoptosis through the mitochondrial apoptosis pathway. Recent evidence has shown that PNAS4 may be involved in the genesis of some cancers and could be a potential candidate for lung cancer therapy alone or in combination with gemcitabine. At least two isoforms of PNAS4 are known to exist.

Pyruvic acid is an intermediate in sugar metabolism and in enzymatic carbohydrate degradation (alcoholic fermentation) where it is converted to acetaldehyde and CO2 by carboxylase. In muscle, pyruvic acid (derived from glycogen) is reduced to lactic acid during exertion, which is reoxidized and partially retransformed to glycogen during rest. It improves coliform recovery when present in culture medium. It is involved in a metabolic regulatory pathway activated by mitochondrial oxidants. Pyruvate is involved in respiratory regulation in plants by interacting with alternative oxidase at a conserved cysteine residue. It may help prevent hydrogen peroxide mediated cell death.
Sodium pyruvate is utilized as a component in culture broth and media. Sodium pyruvate is used in Wallen fermentation medium to enhance the conversion of oleic acid to 10-ketostearic acid by Bacillus sphaericus. Sodium pyruvate has also been used to establish stably transfected human B cell lines.
Sodium Pyruvate has shown antioxidant properties and protective effects against oxygen radicals. Pyruvate is produced as part of glycolysis and is an intermediate in many metabolic pathways. It can be converted into acetyl CoA and enter the TCA Cycle.

Glutathione is the major low molecular weight thiol compound of the living plant or animal cell. It is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. The sulfhydryl (thiol) group (SH) of cysteine serves as a proton donor and is responsible for the biological activity of glutathione.
Glutathione suppresses human immunodeficiency virus expression in chronically infected monocytic cells. It is a useful tripeptide involved in many aspects of metabolism, including transport of g-glutanyl amino acids and reductive cleavage of disulfide bonds.
Endogenous antioxidant that plays a major role in reducing reactive oxygen species formed during cellular metabolism and the respiratory burst. Glutathione-S-transferase catalyzes the formation of glutathione thioethers with xenobiotics, leukotrienes, and other molecules that have an electrophilic center. Glutathione also forms disulfide bonds with cysteine residues in proteins. Via these mechanisms, it can have the paradoxical effect of reducing the efficacy of anti-cancer agents.
