This gene encodes an alpha-1 subunit of a voltage-dependent calcium channel. Calcium channels mediate the influx of calcium ions into the cell upon membrane polarization. The alpha-1 subunit consists of 24 transmembrane segments and forms the pore through which ions pass into the cell. The calcium channel consists of a complex of alpha-1, alpha-2/delta, beta, and gamma subunits in a 1:1:1:1 ratio. There are multiple isoforms of each of these proteins, either encoded by different genes or the result of alternative splicing of transcripts. PCDH17 is a 1,159 amino acid single-pass type I membrane protein that contains six cadherin domains. Expressed as multiple alternatively spliced isoforms, PCDH17 is thought to function as a calcium-dependent cell adhesion protein that may play a role in establishing cell-cell connections within brain tissue. Syk (72 kDa) is a non receptor protein tyrosine kinase that plays an important role in immune receptor signal transduction and is implicated in endothelial cell functions, including cell growth and migration. SYK is a positive effector of BCR stimulated responses. It couples the B cell antigen receptor (BCR) to the mobilization of calcium ions either through a phosphoinositide 3 kinase dependent pathway, when not phosphorylated on tyrosines of the linker region, or through a phospholipase C gamma dependent pathway, when phosphorylated on Tyr 342 and Tyr 346. Thus the differential phosphorylation of SYK can determine the pathway by which BCR is coupled to the regulation of intracellular calcium ions. RecQ helicases are implicated in DNA recombination and repair helicases and are critical in genome maintenance. RecQL5 is one of five known RecQ homologues in humans, and defects in three of these (BLM, WRN and RecQ4)are associated with autosomal recessive disorders that involve genomic instability and a predisposition to cancer. The exact role of ReqL5 is unclear. It has been shown in mouse embryonic stem (ES) cells that mutations in the Recql5 genes result in a significant increase in the frequency of sister chromatid exchange (SCE), whereas deleting Recql5 lead to an even higher frequency of SCE. There are three different RecQL5 isoforms - alpha, beta and gamma, produced by alternative splicing.
