Adducins are a family of cytoskeleton proteins encoded by three genes (alpha, beta, gamma). Adducin is a protein associated with the inner leaflet of the plasma membrane and is one of the proteins localised at the spectrin-actin junction of the membrane skeleton. The cortical actin cytoskeletal network is lost during apoptosis and Adducins are central in the cortical actin network organisation. Adducin Alpha is a cytoskeletal protein involved with sodium-pump activity in the renal tubule and is associated with hypertension. The expression of Adducin Alpha and Adducin Gamma is ubiquitous in contrast to the restricted expression of Adducin Beta. Adducin Beta is expressed at high levels in brain and hematopoietic tissues, such as bone marrow, in humans, and in spleen in mice.

Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. Expression of MVP predicts a poor response to chemotherapy.

The PHEX a 749 amino acid protein that putatively consists of an intracellular, transmembrane and extracellular domain. PHEX mutations have been observed in 60-80% of hypophosphatemic rickets patients. The PHEX protein, which is a single-pass membrane protein, is also designated HYP, X-linked hypophosphatemia protein or metalloendopeptidase homolog PEX. PHEX plays an active role in bone and dentin mineralization and renal phosphate re-absorption. X-linked hypophosphatemic rickets, also designated HYP, is an X-linked dominant disorder characterized by impaired phosphate uptake in the kidney, which is likely to be caused by abnormal regulation of sodium phosphate cotransport in the proximal tubules. Clinical manifestations include skeletal deformities, growth failure, craniosynostosis, paravertebral calcifications, pseudofractures in lower extremities, and muscular hypotonia with onset in early childhood.

The sodium-coupled neutral amino acid transporters (SNAT) of the SLC38 gene family include System A subtypes SNAT1, SNAT2 and SNAT4 and System N subtypes SNAT3 and SNAT5. The SLC38 transporters are essential for the uptake of nutrients, energy production, metabolism, detoxification, and the cycling of neurotransmitters. SNAT2, also designated ATA2, PRO1068 and SAT2 is encoded by the human gene SLC38A2. The functional role of SNAT2 in the nervous system is unclear. Protein expression is notably enriched in the spinal cord and brain stem nuclei of the auditory system. System A transport proteins are also present in placental tissue. These SNAT proteins may play a significant role in fetal development and inhibition of the transport system has been associated with fetal growth retardation.

CBG is a monomeric enzyme involved in the absorption and metabolism of flavonoid glucosides. CBG is found predominately in the liver, but is also located in tissues such as spleen, small intestine and kidney. Through its catalytic activity, CBG is able to hydrolyze a variety of glycosides including phytoestrogens, cyanogens, and flavonols. Although its catalytic activity extends to many dietary flavonoids, CBG has increased specificity for hydrophobic aglycones such as beta-D-glucoside and beta-D-galactoside. Hydrolysis is inhibited by sodium taurocholate and glucosyl-sphingosine, both of which regulate CBG enzymatic activity. Deficiencies in CBG have been implicated in Gaucher’s disease, a lysosomal storage disease that causes a build up of fatty material in the spleen, liver, lung and kidneys.

Neuronal nitric oxide synthase (nNOS), which lacks the T/SXV motif but which has its own PDZ domain, has been shown to associate with PSD 93 in vitro through a pseudo-homotypic PDZ-PDZ interaction.
