Uteroglobin belongs to the family of secretoglobins and is a secreted protein product of nonciliated bronchiolar Clara cells. There is convincing data suggesting it has phospholipase A2 inhibitory activity, as well as, a number of other immunomodulatory features including inhibition of interferon gamma signalling and Th1 vs. Th2 lymphocyte regulation. It was proposed as a potential peripheral marker of respiratory epithelial injury and bronchial dysfunction. Clara Cell Protein 16 concentrations have been determined in both serum and bronchoalveolar lavage fluid in numerous studies since 1994. In serum, its increase is associated with age, asbestos, nitrogen chloride and ozone exposure, sarcoidosis and high PEEP ventilation. Decreased serum CC16 levels are found after pulmonary resection, in silica-exposed workers, smokers and in asthma. Decreased CC16 concentrations were also found in the amniotic fluid of fetuses suffering from pulmonary hypoplasia caused by various mechanisms (diaphragmatic hernia, diabetic fetopathy, Turner and Down syndrome). In pleural effusions, the CC16 concentration appears to be associated with its diffusion from the lung as evidenced by high CC16 levels in cardiac pleural congestion.

Human papillomaviruses, particularly type 16 (designated HPV16), infect the genital tract and may lead to cervical cancer. Protection against HPV16 is thought to be provided by neutralizing directed to the major caspid protein L1 of HPV16. HPV16 L1 forms the pentameric assembly unit of the viral shell, and the binding of HPV16 L1 to the cell surface without the involvement of minor capsid protein L2 is believed to be the first step of HPV16 infection. The L1-binding domain located near the C-terminus of L2 binds L1 prior to completion of capsid assembly and is required for efficient encapsidation of the viral genome. In addition, the C-terminus of L1 is necessary for both DNA binding and DNA packaging. Expression of the late gene L1 is restricted to the upper layers of the infected epithelium. HPV16 L1 is able to package unrelated plasmid DNA in vitro and deliver the foreign DNA to eukaryotic cells with the subsequent expression of the encoded gene. L1 shows a diffuse nuclear distribution whereas L2 is localized to punctate nuclear regions identified as promonocytic leukemia protein oncogenic domains (PODs). Coexpression of L1 and L2 induces a relocalization of L1 into the PODs.

Mouse interleukin-16(IL-16) is a single chain non-glycosylated polypeptide. IL-16 is widely expressed in human tissues including spleen, thymus, lymph nodes, peripheral leukocytes, bone marrow and cerebellum. IL-16 plays an important role instimulating a migratory response in CD4+ lymphocytes, monocytes, and eosinophils,inducing T-lymphocyte expression of interleukin 2 receptor.It was originally identified as a CD8+ T cell-derived chemoattractant for CD4+ cells. In addition to its chemotactic properties, IL-16 has also been shown to suppress HIV-1 replication in vitro and appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. It may act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells.

This MAb recognizes a protein of 56 kDa, identified as cytokeratin 6 (CK6). In humans, multiple isoforms of Cytokeratin 6 (6A-6F), encoded by several highly homologous genes, have distinct tissue expression patterns, and Cytokeratin 6A is the dominant form in epithelial tissue. The gene encoding human Cytokeratin 6A maps to chromosome 12q13, and mutations in this gene are linked to several inheritable hair and skin pathologies. Keratins 6 and 16 are expressed in keratinocytes, which are undergoing rapid turnover in the suprabasal region (also known as hyper-proliferation-related keratins). Keratin 6 is found in hair follicles, suprabasal cells of a variety of internal stratified epithelia, in epidermis, in both normal and hyper-proliferative situations. Epidermal injury results in activation of keratinocytes, which express CK6 and CK16. CK6 is strongly expressed in about 75% of head and neck squamous cell carcinomas. Expression of CK6 is particularly associated with differentiation.
