DTYMK is a 212 amino acid protein that belongs to the thymidylate kinase family and is involved in pyrimidine metabolism. Specifically, DTYMK catalyzes the ATP-dependent conversion of dTMP (deoxythymidine monophosphate) to dTDP (deoxythymidine diphosphate), which then functions as one of the four nucleotides in DNA. Via its role in the catalytic creation of dTDP, DTYMK plays an important role in the pathway of DNA synthesis and is thought to be involved in cell cycle progression and cell growth. DTYMK expression levels peak during the S phase (synthesis phase) of the cell cycle, further supporting the role of DTYMK in DNA synthesis.

GIOT-1 is a 563 amino acid protein belonging to the Krüppel C2H2-type zinc-finger protein family. Localized to the nucleus, GIOT-1 is widely expressed in tissues, with highest levels in liver, kidney, small intestine, pancreas and thymus. GIOT-1 contains 12 C2H2-type zinc fincers and one KRAB domain. Because the KRAB domain functions as a transcriptional repressor when attached to the template DNA, GIOT-1 is thought to be involved in transcriptional regulation. The gene encoding GIOT-1 is localized to chromosome 19q13.12 and two isoforms of GIOT-1 exist as a result of alternative splicing events.

DUSPs are a heterogeneous group of protein phosphatases that can dephosphorylate both phosphotyrosine and phosphoserine/phosphothreonine residues within the one substrate. These phosphatases inactivate their target kinases by dephosphorylating both the phosphoserine/threonine and phosphotyrosine residues. DUSPs are major modulators of critical signalling pathways that are dysregulated in various diseases. They negatively regulate members of the mitogen-activated protein kinase superfamily, which are associated with cellular proliferation and differentiation. DUSP3 is expressed in human tissues including breast and ovarian.DUSP3 shows activity both for tyrosine-protein phosphate and serine-protein phosphate, but displays a strong preference toward phosphotyrosines. mTOR is one of a family of proteins involved in cell cycle progression, DNA recombination, and DNA damage detection.
