NCS-1 (for neuronal calcium sensor-1, also designated frequenin) belongs to a highly conserved family of EF-hand-containing Ca++-binding proteins expressed mainly in neurons. NCS-1 is localised to neuronal cell bodies and axons throughout the brain and spinal cord. It is also expressed in glial cells and in neuroendocrine bovine adrenal chromaffin and PC12 cells. NCS-1 is a regulatory protein involved in Ca++-dependent exocytosis of synaptic vesicles and dense core granules. NCS-1 also functions in the voltage-independent autocrine pathway that negatively regulates non-L-type Ca++ channels.

APPBP2 is a hydrophilic, microtubule binding protein that functions in the trafficking of Beta-Amyloid precursor protein. It is expressed in a variety of cell types and localizes to the cytoplasm. APPBP2 shares homology with kinesin light chain. It consists of a coiled-coil domain, PKC phosphorylation sites, four imperfect C-terminal tandem repeats, eight tetratricopeptide repeats and N- and C-terminal globular structures. APPBP2 recognizes and binds to the basolateral sorting sequence (BaSS) present in the cytoplasmic domain of the Beta-Amyloid precursor protein. In addition, APPBP2 interacts with the androgen receptor and suppresses androgen Signalling.

Bad is a member of the Bcl2 family and acts to promote apoptosis by forming heterodimers with the survival proteins Bcl2 and BclxL, thus preventing them from binding with BAX. Bad is found on the outer mitochondrial membrane and, once phosphorylated in response to growth stimuli, translocates to the cytoplasm. The phosphorylation status of Bad represents a key checkpoint for death or cell survival. JNK-induced phosphorylation of BAD serine 128 promotes the apoptotic role of Bad by opposing the inhibitory effect of growth factor on Bad-mediated apoptosis. Cdc2-induced phosphorylation of Bad serine 128 has an inhibitory effect on its interaction with 14-3-3 proteins. The latter interaction is critical for Bad phosphorylation at serine 155, a site within the BH3 domain that leads to the release of BclxL and the promotion of cell survival. Alternative splicing of this gene results in two transcript variants which encode the same isoform.This antibody recognise the phosphorylation site of human Ser75, mouse Ser111, rat Ser112.

Midline-2 is a 715 amino acid protein encoded by the human gene MID2. Midline-2 belongs to the TRIM/RBCC family and contains two B box-type zinc fingers, one B30.2/SPRY domain, one COS domain, one fibronectin type-III domain and one RING-type zinc finger. Midline-2 is a cytoplasmic protein found as a homodimer or heterodimer with Midline-1. It also interacts with IGBP1 (Lymphocyte signaling protein A4). Dimerization is mediated by the tripartite motif (RBCC; RING- and B box-type zinc fingers and coiled coil domains) and microtubule association is dependent on the C-terminal B30.2 domain. Midline-2 is expressed at low levels in fetal kidney and lung, and in adult prostate, ovary and small intestine.

Because of its high homology to other known VAMPs, its broad tissue distribution, and its subcellular localization, the protein encoded by this gene was shown to be the human equivalent of the rodent cellubrevin. Expressed at high levels in brain and present at lower levels in lung tissue, EPHEXIN interacts with EphA4 and functions as a guanine nucleotide exchange factor (GEF) that is capable of activating Rho A, Rac 1 and Cdc42 and is thought to play a role in axon guidance and growth cone collapse. EPHEXIN is subject to Src-dependent phosphorylation, an event that increases the GEF activity of EPHEXIN toward Rho A. Human EPHEXIN, which exists as multiple alternatively spliced isoforms, shares a high degree of sequence homology with its mouse counterpart, suggesting a conserved role between species.

CSNK1A1L resides in the cytoplasm and participates in the Wnt signaling pathway. By utilizing ATP within its protein kinase domain, CSNK1A1L phosphorylates a large number of proteins.

AGPS is a 658 amino acid enzyme that is required for glycerolipid metabolism and ether lipid biosynthesis. Localized to the inner aspect of the peroxisomal membrane, AGPS is likely part of a heterotrimeric complex that is also composed of GNPAT and a modified form of GNPAT. Containing one FAD-binding PCMH-type domain, AGPS utilizes FAD as a cofactor in the synthesis of alkyl-glycerone 3-phophate and a long-chain acid anion from 1-acteyl-glyerone 3-phosphate and a long-chain alcohol. Defects in the gene encoding AGPS results in rhizomelic chondrodysplasia punctata type 3, a disease characterized by vertebral disorders, severe mental retardation, cutaneous lesions, cataracts and rhizomelic shortening of the humerus and femur.
