Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.

Recognizes a non-histone protein of 36 kDa, which is identified as proliferating cell nuclear antigen (PCNA). It is also known as cyclin or polymerase delta auxiliary protein. Elevated expression of PCNA/cyclin has been shown in the nucleus during late G1 phase immediately before the onset of DNA synthesis, becoming maximal during S-phase and declining during G2 and M phases. This MAb is excellent for multiple applications.

Recognizes a 34 kDa protein, which is identified as cyclin dependent kinase 1 (cdk1) or p34cdc2 protein kinase. cdk1 / p34cdc2 plays a crucial role during cell division and is most active during mitosis. It is predominantly localized in the nucleus. It is a serine/threonine kinase, which is activated by cyclin, presumably by de-phosphorylation of tyrosine residues. Activated cdk1 / p34cdc2 performs specific functions during mitosis, including nuclear envelope breakdown and chromosome condensation.

This antibody recognizes a cell surface glycoprotein of 95/115/135 kDa (depending upon the extent of glycosylation), identified as CD43. 70-90% of T-cell lymphomas and from 22-37% of B-cell lymphomas express CD43. No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma.

It is also the product of proteolytic cleavage, which results in a secreted protein.

Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). CK8 is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Hepatocellular carcinomas are defined by the use of antibodies that recognize only cytokeratin 8 and 18. CK8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Anti-CK8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against CK8, CK18 and CK19). Reportedly, anti-CK8 is useful for the differentiation of lobular (ring-like, perinuclear) from ductal (peripheral-predominant) carcinoma of the breast.

Pax-7 is a protein specifically expressed in cultured satellite cell-derived myoblasts. In situ hybridization reveals that Pax-7 is also expressed in satellite cells residing in adult muscle. A chromosomal aberration in the gene encoding Pax-7 causes rhabdomyosarcoma 2 (RMS2) (also called alveolar rhabdomyosarcoma).
