VPS4A is a member of the AAA protein family (ATPases associated with diverse cellular activities), and is the homolog of the yeast Vps4 protein. In humans, two paralogs of the yeast protein have been identified. Functional studies indicate that both human paralogs associate with the endosomal compartments, and are involved in intracellular protein trafficking, similar to Vps4 protein in yeast. The protein encoded by this gene is a member of the AAA protein family (ATPases associated with diverse cellular activities), and is the homolog of the yeast Vps4 protein. In humans, two paralogs of the yeast protein have been identified. The former share a high degree of aa sequence similarity with each other, and also with yeast Vps4 and mouse Skd1 proteins. The mouse Skd1 (suppressor of K+ transport defect 1) has been shown to be really an yeast Vps4 ortholog. Functional studies indicate that both human paralogs associate with the endosomal compartments, and are involved in intracellular protein trafficking, similar to Vps4 protein in yeast. The gene encoding this paralog has been mapped to chromosome 16; the gene for the other resides on chromosome 18. Publication Note: This RefSeq record includes a subset of the publications that are available for this gene. Please see the Entrez Gene record to access additional publications.

TFAM a mitochondrial transcription factor that is a key activator of mitochondrial transcription as well as a participant in mitochondrial genome replication. Studies in mice have demonstrated that this protein is required to regulate the mitochondrial genome copy number and is essential for embryonic development. A mouse model for Kearns Sayre syndrome was produced when expression of the gene was eliminated by targeted disruption in heart and muscle cells.

DBX2 Antibody: DBX2 is a member of the developing brain homeobox (DBX) protein family, but while the related protein DBX1 is expressed in various regions of the developing brain, DBX2 shows a more restricted pattern of expression in the brain, and is also expressed in some mesenchymal cells such as limb buds and tooth germs. It is thought that DBX1 and DBX2 promote the development of a subset of interneurons, some of which help mediate left-right coordination of locomotor activity. In Xenopus, DBX2 is involved in primary neurogenesis and early neural plate patterning, and is thought to act as a cross-repressive partner of NKX6-2 in the patterning of the ventral neural tube.

BMP15 Antibody: BMP15, or GDF9B is a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily that are involved in embryonic development and adult tissue homeostasis. BMP15 is expressed exclusively in the oocyte. It is an oocyte-specific growth/differentiation factor that may be involved in oocyte maturation and follicular development. Defects in BMP15 are the cause of ovarian dysgenesis type 2. BMP15 could be used as an oogenesis marker to track human amniotic fluid stem cells differentiation into the oocyte-like cells.

S100 calcium binding protein A1 (S100-alpha/ S100A1), The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein may function in stimulation of Ca2+-induced Ca2+ release, inhibition of microtubule assembly, and inhibition of protein kinase C-mediated phosphorylation. Reduced expression of this protein has been implicated in cardiomyopathies.

Ractopamine is a phenolethanolamine beta-adrenoceptor agonist that is used for the improvement of weight gain, carcass leanness and feed efficiency in pigs. It has been known for years that ractopamine enhances growth in meat animals by repartitioning nutrients. This means that the nutrients that the animal consumes in its feed are shifted away from being stored as fat and are instead used in lean tissue production. This shift occurs as the animal reaches the top of its growth curve and lean tissue production drops off and fat deposition increases. Fat levels also appear to decrease, due to an increase in lypolysis (fat breakdown). Using the ractopamine monoclonal antibody, it is possible to detect ractopamine and other agonists in urine, muscle and liver both rapidly and with accuracy.

Rsk1 is a member of a family of 90 kDa ribosomal protein S6 kinases, which includes Rsk1, Rsk2 and Rsk3. These are broadly expressed serine / threonine protein kinases activated in response to mitogenic stimuli, including extracellular signal regulated protein kinases Erk1 and Erk2. Rsk1 is activated by MAPK in vitro and in vivo via phosphorylation. Active Rsks appear to play a major role in transcriptional regulation by translocating to the nucleus and phosphorylating c-Fos and CREB. RSK proteins possess two separate kinase domains, the C terminal kinase domain and the N terminal kinase domain, separated by a linker region containing a hydrophobic motif. RSK is activated by many stimuli including growth factors, phorbol esters, cAMP, heat shock, and irradiation.

RNASEH1 belongs to the RNase H family. It contains 1 RNase H domain. RNASEH1 is an endonuclease that specifically degrades the RNA of RNA-DNA hybrids.

Translation initiation is mediated by specific recognition of the cap structure by eukaryotic translation initiation factor 4F (eIF4F), which is a cap binding protein complex that consists of three subunits: eIF4A, eIF4E and eIF4G. EIF4G2 shares similarity with the C-terminal region of eIF4G that contains the binding sites for eIF4A and eIF3; eIF4G, in addition, contains a binding site for eIF4E at the N-terminus. Unlike eIF4G, which supports cap-dependent and independent translation, EIF4G2 functions as a general repressor of translation by forming translationally inactive complexes. In vitro and in vivo studies indicate that translation of this mRNA initiates exclusively at a non-AUG (GUG) codon. Alternatively spliced transcript variants encoding different isoforms of this gene have been described.
