SPG15 Antibody: Hereditary spastic paraplegias (HSPs) are genetically and phenotypically heterogeneous disorders. Spastic paraplegia with thinning of the corpus callosum (ARHSP-TCC) is a relatively frequent form of complicated hereditary spastic paraplegia in which mental retardation and muscle stiffness at onset are followed by slowly progressive paraparesis and cognitive deterioration. SPG15 is the second gene known to be responsible for ARHSP-TCC in the Italian population. Mutations in this gene are associated with autosomal recessive spastic paraplegia-15. SPG15 encodes a protein containing a FYVE zinc finger binding domain which is thought to target these proteins to membrane lipids through interaction with phospholipids in the membrane. SPG15 mRNA is widely distributed in human tissues, as well as in rat embryos, suggesting a possible role for this protein during embryonic development. SPG15 co-localizes partially with endoplasmic reticulum and endosome markers, suggesting a role in intracellular trafficking. Multiple isoforms of SPG15 are known to exist.

PRDM16 Antibody: PRDM16 is a zinc finger transcription factor and contains an N-terminal PR domain. The reciprocal translocation t(1;3)(p36;q21) occurs in a subset of myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML). This gene is located near the 1p36.3 breakpoint and has been shown to be specifically expressed in the t(1:3)(p36, q21)-positive MDS/AML. The translocation results in the overexpression of a truncated version of this protein that lacks the PR domain, which may play an important role in the pathogenesis of MDS and AML. Recent studies have shown that PRDM16 normally acts as a Smad3 binding protein that may be important for the development of orofacial structures through modulation of the TGF-beta signaling pathway. Other experiments have indicated that PRDM16 controls a bidirectional cell fate switch between skeletal myoblasts and brown fat cells.

ANGPTL4 mainly expressed in endothelial cells (hypoxia-induced). Regulates angiogenesis and modulates tumorgenesis and directly regulates lipid, glucose, and energy metabolism. Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. ANGPTL4 is a protein consisting of an N-terminal coiled-coil domain and a C-terminal fibrinogen-like domain (FLD). Both domains have distinct biological functions. The coiled-coil domain is responsible for the inhibitory effects on lipoprotein lipase (LPL) converting the active form of LPL into an inactive form, and the FLD domain mediates its antiangiogenic functions. The coiled coil and the FLD domains are separated by a short linker that can be cleaved after secretion. ANGPTL4 appears on the cell surface as the full-length form, where it can be released by heparin treatment. ANGPTL4 protein is then proteolytically cleaved by proprotein convertases (PCs), including furin, PC5/6, paired basic amino acid-cleaving enzyme 4, and PC7.

Krueppel-Like Factor 6 (KLF6) belongs to the krueppel C2H2-type zinc-finger protein family. KLF6 contains three C2H2-type zinc fingers and localises in the nucleus. KLF6 expression is highest in the placenta followed by spleen, thymus, prostate, testis, small intestinem and colon. However, it is weakly expressed in the pancreas, lung, liver, heart, and skeletal muscle. KLF6 functions as a transcriptional activator and could play a role in B-cell growth and development. Defects in KLF6 will result in gastric cancer and prostate cancer.

NALP7 has a C-terminal leucine-rich repeat (LRR) region, an N-terminal Pyrin domain (PYD) followed by a NACHT domain, and a NACHT-associated domain. It is expressed in numerous tissues including uterus and ovary, with low levels in heart and brain. NALP7 inhibits caspase-1-dependent interleukin-1 beta secretion and is a feedback regulator of interleukin-1 beta secretion. Defects in the NALP7 gene are known to cause the formation of a hydatidiform mole (HYDM) and reduce the growth of carcinoma cell lines.
