Tumor suppressor. Acts as a dual-specificity protein phosphatase, dephosphorylating tyrosine-, serine- and threonine-phosphorylated proteins. Also acts as a lipid phosphatase, removing the phosphate in the D3 position of the inositol ring from phosphatidylinositol 3, 4, 5-trisphosphate, phosphatidylinositol 3, 4-diphosphate, phosphatidylinositol 3-phosphate and inositol 1, 3, 4, 5-tetrakisphosphate with order of substrate preference in vitro PtdIns(3, 4, 5)P3 > PtdIns(3, 4)P2 > PtdIns3P > Ins(1, 3, 4, 5)P4. The lipid phosphatase activity is critical for its tumor suppressor function. Antagonizes the PI3K-AKT/PKB signaling pathway by dephosphorylating phosphoinositides and thereby modulating cell cycle progression and cell survival. The unphosphorylated form cooperates with AIP1 to suppress AKT1 activation. Dephosphorylates tyrosine-phosphorylated focal adhesion kinase and inhibits cell migration and integrin-mediated cell spreading and focal adhesion formation. May be a negative regulator of insulin signaling and glucose metabolism in adipose tissue.

Aipl1 Antibody: Aipl1 was initially identified as a protein implicated in Leber congenital amaurosis (LCA), an autosomal recessive disorder thought to be caused by the abnormal development of photoreceptors. Aipl1 is a tetratricopeptide repeat protein that is highly homologous to ARA9, a protein involved in the HSP90-mediated nuclear translocation and transactivation of the aryl hydrocarbon receptor. Aipl1 has also been found to function as part of a chaperone heterocomplex, interacting with Hsp90 and Hsp70. Aipl1 also associates with the cell cycle regulator NUB1. It is thought that Aipl1 cooperates with Hsp70 but not Hsp90 to suppress the formation of NUB1 inclusions, and these interactions are necessary in the normal photoreceptor maturation, as mutations that lead to LCA also compromise the interactions with the Hsp chaperones. At least three isoforms of Aipl1 are known to exist.

LGP2 Antibody: Anti-viral innate immune responses are triggered by pathogen-associated molecular patters (such as the accumulation of intracellular nucleic acids resulting from virus infections) and represent the first line of defense against numerous infectious organisms. The Toll-like receptors (TLR) 3, 7, 8, and 9 are expressed in immune cells and function as transmembrane pattern recognition receptors to detect foreign nucleic acids. Other proteins that play similar roles, such as RIG-1 and MDA5 are members of a CARD-helicase family and are expressed in the cytoplasm. A third protein, LGP2, is similar to RIG-1 and MDA5, except for lacking a homologous CARD domain. It is thought to act as an element of negative-feedback regulation of intracellular antiviral signaling. At least four isoforms of LGP2 are known to exist.

CRTH2 Antibody: The chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTH2) is a recently identified receptor for the prostaglandin D2 (PGD2) in addition to the classic prostaglandin D receptor. CRTH2 is expressed on Th2 cells and eosinophils and mediates chemotaxis of these cells to PGD2 and is thus thought to be a key receptor mediating eosinophil and Th2 recruitment during allergic responses. However, CRTH2-null mice showed enhanced eosinophil recruitment into the lung consistent with observations that the CRTH2-null mice produced significantly higher amounts of interleukin-5 (IL-5) and IL-3. This suggests that CRTH2 plays a nonredundant role in restricting eosinophilia and allergic response in vivo. At least two different isoforms of CRTH2 are known to exist.

ERBB2 is widely expressed in epithelial cells, and amplification and/or overexpression of ErbB2 has been reported associated with malignancy and a poor prognosis in numerous carcinomas, including breast, prostate and ovarian cancers. FMS-like tyrosine kinase 3 ligand (Flt-3 Ligand) is also known as FL, Flt3L and FLT3LG, is an α-helical cytokine that promotes the differentiation of multiple hematopoietic cell lineages. FLT3LG is expressed as a noncovalentlylinked dimer by T cells and bone marrow and thymic fibroblasts. Each 36 kDa chain carries approximately 12 kDa of N- and O- linked carbohydrates. FLT3LG is structurally homologous to stem cell factor (SCF) and colony stimulating facor 1 (CSF-1). FLT3LG acts as a growth factor that increases the number of immune cells by activating the hematopoietic progenitors. It also induces the mobilisation of the hematopoietic progenitors and stem cells in vivo which may help the system to kill cancer cells. FLT3LG induces the expansion of monocytes and immature dendritic cells as well as early B cell lineage differentiation. FLT3LG cooperates with IL2, IL6, IL7, and IL15 to induce NK cell development and with IL3, IL7 and IL11 to induce terminal B cell maturation. Animal studies also show FLT3LG to reduce the severity of experimentally induced allergic inflammation. FLT3LG is crucial for steady-state pDC and cDC development. A lack of FLT3L results in low levels of DCs.

Neurturin Antibody: Glial cell line-derived neurotrophic factor (GDNF) plays key roles in the control of vertebrate neuron survival and differentiation. A novel neurotrophic factor was recently cloned from human and mouse and designated neurturin. Physiological responses to neurturin (NTN) require the presence of receptor tyrosine kinase RET and a novel glycosylphosphatidylinositol linked receptor NTNRalpha. The cDNAs encoding NTNRalpha from human, rat, chicken, and mouse have been cloned recently and termed GDNFR beta , Ret ligand 2 (RETL2) or TGF-beta-related neurotrophic factor receptor 2 (TrnR2) and nominated as GFR alpha -2 recently. NTN binds to and forms a complex with GFR alpha -2 and the Ret PTK and activates the RET receptor tyrosine kinase pathway. Both NTN and GDNF can activate the MAP kinase and phosphatidylinositol 3-kinase pathways and play a critical role in the development of many neuronal populations. Neurturin and GDNF define a new family of neurotrophic factors.

Apaf-1 Monoclonal Antibody: Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. The mammalian homologous of the key cell death gene CED-4 in C. elegans was identified recently from human and mouse and designated Apaf1 for apoptosis protease-activating factor 1. Apaf1 binds to cytochrome c (Apaf2) and caspase-9 (Apaf3), which leads to caspase-9 activation. Activated caspase-9 in turn cleaves and activates caspase-3 that is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis. Apaf1 can also associate with caspase-4 and caspase-8. Apaf1 transcript is ubiquitously expressed in human tissues.

CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumor growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. PD-L1 thus is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.
