It recognises an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This mAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. This mAb is highly useful in distinguishing ovarian carcinomas (keratin 7+) from colon carcinomas (keratin 7-).

Recognises a 40-43 kDa transmembrane epithelial glycoprotein, identified as epithelial specific antigen (ESA), or epithelial cellular adhesion molecule (Ep-CAM). EpCAM is expressed on baso-lateral cell surface in most simple epithelia and a vast majority of carcinomas. This antibody has been used to distinguish adenocarcinoma from pleural mesothelioma and hepatocellular carcinoma. It is also useful in distinguishing serous carcinomas of the ovary from mesothelioma. This epithelial antigen plays an important role as a tumor-cell marker in lymph nodes from patients with esophageal carcinoma otherwise classified as node-negative. Epithelial antigen has also been suggested as a discriminator between basal cell and baso-squamous carcinomas, and squamous cell carcinoma of the skin.

It recognises an intra-cytoplasmic antigen, which shows a very high degree of specificity for plasma cells. This antigen is present in normal as well as neoplastic plasma cells. Plasma cells, which are large lymphocytes derived from an antigen-specific B cell, secrete antibodies and are responsible for humoral immunity. Plasma cells differentiate from B cells upon stimulation by CD4+ lymphocytes. The B cell acts as an antigen-presenting cell (APC), consuming an offending pathogen, which is taken up by the B cell by phagocytosis and broken down within proteosomes. Plasma cells contain basophilic cytoplasm; their nucleus contains heterochromatin organized in a characteristic cartwheel arrangement. This mAb superbly recognises normal and neoplastic plasma cells in routine formalin-fixed, paraffin-embedded tissue sections. It is of potential value in identifying myeloma or plasmacytoma in bone marrow or other tissues. It also helps differentiate lympho-plasmacytoid lymphoma from lymphocytic and follicular lymphoma.

The UC10-4F10-11 monoclonal antibody specifically reacts with the mouse Cytotoxic T-Lymphocyte Antigen-4 (CTLA-4), also known as CD152. It is a protein with a structure similar to CD28 regarding the genomic organization, amino acid sequence, and structure. CTLA-4 is expressed on activated T cells and CD25+/CD4+ Treg lymphocytes and binds the members of the B7 family, B7-1 (CD80) and B7-2 (CD86), with higher affinity than CD28. CD28 seems to provide opposing signal to T lymphocytes, while CD152 inhibits the T lymphocytes progression to an activated state and their proliferation, CD28 is a costimulator. The mouse UC10 -4F10-11 monoclonal antibody does not cross-react with the rat leukocytes.
