The CD28.2 monoclonal antibody specifically binds with the human 44 kDa homodimeric trans-membrane glycoprotein CD28, expressed on the surface of most mature T lymphocytes, plasma cells, and thymocytes. CD28 is a ligand for B7-1 (CD80) and B7-2 (CD86), a co-stimulator of T lymphocytes, and enhances the interaction between T and B lymphocytes. It has been reported that the T lymphocytes stimulation to produce IL-2 depends on the monoclonal antibody involved, which suggests that the CD28 molecule presents some subregions with distinct functions. The CD28.2 antibody induces Ca2+ influx in Jurkat T lymphocytes. Other studies have shown that CD28 is involved in the signal transduction.

F4/80 is a transmembrane protein used as a marker of macrophages, although it is also expressed on Kupffer and Langerhans cells. SGKL/SGK3, also known as CISK for cytokine-independent survival kinase, is a SGK type protein kinase. The protein contains a lipid-binding phox homology (PX) domain, which is required for targeting SGKL/SGK3 protein to the endosomal compartment. SGKL/SGK3 functions downstream of the phosphoinositide 3-kinase (PI3K) cascade and has been implicated in cell survival. The protein has been shown to be phosphorylated and activated by 3-phosphoinositide-dependent protein kinase 1 (PDK1), and to a lesser extent insulin-like growth factor-1, in vitro. Activated SGKL/SGK3 has been shown to phosphorylate glycogen synthase kinase 3 beta (GSK-3beta) in vitro.

This mAb reacts with a 200 kDa protein, identified as heavy subunit of neurofilaments. Neurofilaments make up the main structural elements of axons and dendrites and are found in neurons, peripheral nerves, and sympathetic ganglion cells. Neurofilaments consist of three major subunits with molecular weights of 68 kDa (NF-L), 160 kDa (NF-M) and 200 kDa (NF-H). Anti-neurofilament stains a number of neural, neuroendocrine, and endocrine tumors. Neuromas, ganglioneuromas, gangliogliomas, ganglioneuroblastomas, and neuroblastomas stain positively for anti-neurofilament. Neurofilaments are also present in paragangliomas as well as adrenal and extra-adrenal pheochromocytomas. Carcinoids, neuroendocrine carcinomas of the skin, and cell carcinomas of the lung also express neurofilament.

Integrin alpha-5 joins with Integrin- beta 1 to form a fibronectin and laminin receptor which recognizes the sequence R-G-D in its ligands. It is expressed on fibroblasts, endothelial cells, peripheral T cells and platelets. Integrin alpha-5 undergoes post-translational cleavage in the extracellular domain to yield disulfide-linked light and heavy chains. In addition to adhesion, ITGA5 participates in cell-surface mediated signalling.
