Fructose Bisphosphate Aldolase A (ALDOA) belongs to the class I fructose-bisphosphate aldolase family. ALDOA is a glycolytic enzyme that catalyses the reversible conversion of fructose-1,6-bisphosphate to glyceraldehyde 3-phosphate and dihydroxyacetone phosphate. In vertebrates, three forms of this ubiquitous glycolytic enzyme are found, Aldolase A in muscle, Aldolase B in liver and aldolase C in brain. Aldolase A Interacts with SNX9 and WAS. Aldolase A deficiency has been associated with myopathy and hemolytic anemia. In addition, Aldolase A plays an important role in glycolysis and gluconeogenesis; it may also act as a scaffolding protein.

Insulin like growth factor binding protein 3 (IGF-BP3) is a member of the superfamily of insulin like growth factor (IGF) binding proteins which include six high affinity IGF binding proteins (IGF-BP) and at least four low affinity binding proteins referred to as IGF-BP related proteins (IGFBPrP). The IGF-BP members are cysteine rich proteins with conserved cysteine residues clustered in the amino terminal and the carboxy terminal regions of the molecule. The cDNA sequence encoding the mature human IGF-BP3 is fused to the signal peptide of CD33. Human IGF-BP3 is the major IGF binding protein in plasma where it exists in a ternary complex with IGFI or IGFII and an acid labile subunit.

Glial Cell Line-Derived Neurotrophic Factor Family Receptor alpha-1 (GDNFR alpha1) is a glycosylphosphatidylinositol (GPI) linked cell surface protein belonging to GDNF-family receptor alpha subtype which consists of at least four members. GFR alpha1and GFR alpha2 are the cognate co-receptor for the neurotrophic factor neurturin mediating the NRTN-induced autophosphorylation and activation of the RET tyrosine kinase receptor. Soluble GFR alphas released enzymatically from the cell surface by phosphatidylinositol phospholipase C, as well as recombinantly produced soluble GFR alpha1, can also bind with high affinity to GDNF and trigger the activation of Ret tyrosine kinase. Human GFR alpha1 shares 93% amino acid identity with mouse GFR alpha1.The expression of the various GFR alphas are differentially regulated in the central and peripheral nervous system, suggesting complementary roles for the GFR alphas in mediating the activities of the GDNF family of neurotrophic factors.

The transcription factor Forkhead-box A1 (FOXA1), also known as hepatocyte nuclear factor 3-alpha, is a member of the FOX class of transcription factors. HNF-1 (alpha and beta), HNF-3 (alpha, beta and gamma), HNF-4 (alpha and gamma), and HNF-6 compose, in part, a homeoprotein family designated the hepatocyte nuclear factor family. The various HNF-1 isoforms regulate transcription of genes in the liver as well as in other tissues such as kidney, small intestine and thymus. FOXA1 is expressed in normal breast ductal epithelium and other epithelium in different organs, such as lung, pancreas, bladder, prostate, and colon. Recently, FOXA1 has been shown to be a major determinant of estrogen-ER activity and endocrine response in breast cancer cells. FOXA1 expression correlates with estrogen receptor (ER)-positivity, especially in luminal subtype A breast cancers, which is associated with favorable prognosis. FOXA1 is useful in the sub-classification of breast carcinomas.

Legumain is a lysosomal cysteine protease which is a member of the peptidase C13 family. Though it is found in many tissues, it is highly expressed in the kidney, heart, and placenta. Legumain has a strict specificity for hydrolysis of asparaginyl bonds and can also cleave aspartyl bonds slowly, especially under acidic conditions. Over-expression Legumain in tumors is significant for invasion and metastasis. Secreted Protein Acidic and Rich in Cysteine (SPARC) is a secreted, evolutionarily conserved collagen-binding glycoprotein and belongs to the SPARC family. SPARC has 286 amino acids and contains an EF-hand in C-termina domain, a follistatin-like domain with Kazal-like sequences. There are two calcium binding sites, one binds 5 - 8 Ca2+ with a low affinity and other on an EF-hand loop that binds a Ca2+ ion with a high affinity. It is highly expressed in tissues undergoing morphogenesis, remodeling and wound repair. SPARC regulate cell growth through interactions with the extracellular matrix (ECM) and cytokines. SPARC bind to numerous proteins of the ECM, affect ECM protein expression, influence cellular adhesion and migration, and modulate growth factor-induced cell proliferation and angiogenesis. SPARC also binds several types of collagen, albumin, thrombospondin, PDGF and cell membranes.
