The MAb11 monoclonal antibody specifically reacts with human tumor necrosis factor alpha (TNF-alpha), a 157 amino acid non-glycosylated protein. TNF-alpha is a pleiotropic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF family of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, auto immune diseases, rheumatoid arthritis, inflammation, and diabetes. The MAB11 antibody was generated from recombinant human TNF and is reported to be a neutralizing antibody.

IL-21 Receptor Antibody: A novel cytokine related to IL2 and IL15 was recently identified and designated IL-21. The receptor for IL-21 (IL-21R, also termed NILR for novel Interleukin receptor) is a new member of the class I cytokine receptor family. IL-21R forms a complex with the common cytokine receptor gamma chain, gammac, and mediates IL-21 signaling. Both IL-21R and the gc are necessary for the IL-21 function. IL-21 and its receptor activate JAK-STAT signaling pathway. IL-21R is expressed in spleen, thymus, natural killer (NK), T and B cell lines. IL-21 plays a role in the proliferation and maturation of NK, B and T cell populations.

The RB6-8C5 monoclonal antibody reacts with the mouse Ly-6G (also known as Gr-1). The Ly-6G protein is a myeloid differentiation antigen of 21 to 25 kDa, expressed in a regulated manner by the myeloid lineage in the bone marrow, where the level of antigen expression is correlated with the granulocyte maturation and differentiation. In the bone marrow, the antigen is not expressed by the erythroid cells. From the peripheric cells, RB6-8C5 binds with monocytes, neutrophils and eosinophils.As a marker for the mouse monocytes, macrophages and granulocytes, the RB6-8C5 antibody is usually combined with M1/70, a macrophage labeling antibody (Anti-CD11b), for phenotypic analysis.

A disulphide-linked heterodimer consisting of mb-1 (or CD79a) and B29 (or CD79b) polypeptides, CD79 is non-covalently associated with membrane-bound immunoglobulins on B cells. This complex of mb-1 and B29 polypeptides and immunoglobulin constitute the B cell Ag receptor. CD79a first appears at pre B cell stage, early in maturation, and persists until the plasma cell stage where it is found as an intracellular component. CD79a is found in the majority of acute leukemias of precursor B cell type, in B cell lines, B cell lymphomas, and in some myelomas. It is not present in myeloid or T cell lines. Anti-CD79a is generally used to complement anti-CD20 especially for mature B-cell lymphomas after treatment with RituximAb (anti-CD20). This antibody will stain many of the same lymphomas as anti-CD20, but also is more likely to stain B-lymphoblastic lymphoma/leukemia than is anti-CD20. Anti-CD79a also stains more cases of plasma cell myeloma and occasionally some types of endothelial cells as well.

BAT1 is a member of the DEAD protein family of ATP-dependent RNA helicases. Members of this family are involved in a number of cellular functions including initiation of translation, RNA splicing, and ribosome assembly. A cluster of genes, BAT1-BAT5, has been localized in the vicinity of the genes for TNF alpha and TNF beta. These genes are all within the human major histocompatibility complex class III region. This protein is a negative regulator of inflammation. It is also thought to be a translation initiation factor. This gene is a strong candidate gene for rheumatoid arthritis. There are multiple alternatively spliced transcript variants known for this gene but only two have been fully described. Both of these variants encode the same isoform. This gene has been found to have multiple polyadenylation sites.
