B7H4 is a B7 family member that negatively regulates T cell immunity by inhibiting of T cell proliferation, cytokine production, and cell cycle progression. In vitro, B7H4 inhibits CD4+ and CD8+ T cell proliferation, cytokine production, and generation of alloreactive cytotoxic T-cells (CTLs). In vivo, blockade of endogenous B7-H4 by specific monoclonal antibody promotes T cell responses. B7-H4 ia an important negative regulator of innate immunity through growth inhibition of neutrophils. B7-H4 is expressed on some tumour cancer cells. The role of B7-H4 in tumour progression may be to transform precancerous cells and then protect them from immunosurveillance.

Mouse Programmed cell death 1 ligand 1(Cd274，PD-L1), is a member of the growing B7 family of immune proteins.It involved in the costimulatory signal essential for T-cell proliferation and IFNG production in a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation by blocking cell cycle progression and cytokine production.B7-H1 has been identified as one of two ligands for programmed death1 (PD1), a member of the CD28 family of immunoreceptors. B7-H1 is constitutively expressed in several organs such as heart, skeletal muscle B7-H1 expression is upregulated in a small fraction of activated T and B cells and a much larger fraction of activated monocytes. The costimulatory function of B7-H1 is critical for enhancing maturation and differentiation of T-cells in lymphoid organs.B7-H1 expression is also induced in dendritic cells and keratinocytes after IFN gamma stimulation. Interaction of B7-H1 with PD1 results in inhibition of TCR-mediated proliferation and cytokine production. The B7-H1:PD1 pathway is involved in the negative regulation of some immune responses and may play an important role in the regulation of peripheral tolerance.

Mannan-Binding Lectin Serine Protease 1 (MASP-1) belongs to the peptidase S1 family. MASP1 contains two CUB domains, one EGF-like domain, one peptidase S1 domain and two Sushi (CCP/SCR) domains. MASP1 is primarily expressed in liver. MASP1 involved in the lectin pathway of the complement, performs a key role in innate immunity by recognizing pathogens through patterns of sugar moieties and neutralizing them. MASP1 is synthesized as a zymogen and activated when it complexes with the pathogen recognition molecules of lectin pathway, the mannose-binding lectin and the ficolins. MASP1 is not directly involved in complement activation but may act as an amplifier of complement activation by cleaving complement C2 or by activating another complement serine protease, MASP2. MASP1 is also able to cleave fibrinogen and factor XIII and may may be involved in coagulation. MASP1 is inhibited by SERPING1 and A2M.
