Interleukin-6 receptor subunit beta (IL6ST) is also known as IL-6 receptor subunit beta, IL-6R subunit beta, IL-6R-beta, IL-6RB, Interleukin-6 signal transducer, Membrane glycoprotein 130 (gp130), CD130, Oncostatin-M receptor subunit alpha and Il6st,which is single-pass type I membrane protein. IL6ST /gp130 /CD130 can be found in tissues such as brain, heart, thymus, spleen, kidney, lung and liver and found in all the cell lines tested except BaF-B03.The expression of IL-6ST /gp130 is not restricted to IL6-responsive cells. The receptor systems for IL6, LIF, OSM, CNTF, IL11, CTF1 and BSF3 can utilize gp130 for initiating signal transmission. IL6ST /CD130 can bind to IL6 /IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduce the signal. IL6ST /GP130 does not bind IL6 and may have a role in embryonic development.

Programmed cell death 1 ligand 1 (PD-L1) is also known as cluster of differentiation (CD274) or B7 homolog 1 (B7-H1), is a member of the growing B7 family of immune molecules and is involved in the regulation of cellular and humoral immune responses. B7-H1 is a cell surface immunoglobulin superfamily with two Ig-like domains within the extracellular region and a short cytoplasmic domain. PD-L1 is highly expressed in the heart, skeletal muscle, placenta and lung and weakly expressed in the thymus, spleen, kidney and liver. PD-L1 is expressed on activated T-cells, B-cells, dendritic cells, keratinocytes and monocytes. PD-L1 is up-regulated on T- and B-cells, dendritic cells, keratinocytes and monocytes after LPS and IFNG activation and up-regulated in B-cells activated by surface Ig cross-linking. PD-L1 involve in the costimulatory signal, essential for T-cell proliferation and production of IL10 and IFNG, in an IL2-dependent and a PDCD1-independent manner.

Mouse SDC4 is a ubiquitous transmembrane proteoglycan which belongs to the syndecan proteoglycan family. SDC4 is a cell surface proteoglycan that bears heparan sulphate. The four vertebrate syndecans, Syndecan-1 through -4, have similar short cytoplasmic domains and extracellular portions that diverge, except for HS attachment sites. Structurally diverse side chains add considerably to the size of the core proteins and serve as binding sites for growth factors, cytokines, and extracellular matrix proteins. Syndecans are present as homodimers or multimers, and are often expressed in developmental and cell type-specific patterns. It is expressed highly in liver, kidney and lung. SDC4 localises to the focal adhesions of adherent cells and binds to a range of extracellular ligands, including growth factors and extracellular-matrix proteins. Through its extracellular domain, syndecan-4 cooperates with adhesion molecules and binds matrix components relevant for cell migration. As a heparan sulphate proteoglycan, SDC4 works as a coreceptor for various growth factors. Syn4 deficiency limits neointimal formation after vascular injury by regulating vascular smooth muscle cells (VSMCs) proliferation and vascular progenitor cells (VPCs) mobilisation. SDC4 have an array of functions including regulating cell growth, differentiation, and adhesion.

ANGPTL4 (Angiopoietin-like protein 4) mainly expressed in endothelial cells (hypoxia-induced). Regulates angiogenesis and modulates tumorigenesis and directly regulates lipid, glucose, and energy metabolism. Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. ANGPTL4 is a protein consisting of an N-terminal coiled-coil domain and a C-terminal fibrinogen-like domain (FLD). Both domains have distinct biological functions. The coiled-coil domain is responsible for the inhibitory effects on lipoprotein lipase (LPL) converting the active form of LPL into an inactive form, and the FLD domain mediates its antiangiogenic functions. The coiled coil and the FLD domains are separated by a short linker that can be cleaved after secretion. ANGPTL4 appears on the cell surface as the full-length form, where it can be released by heparin treatment. ANGPTL4 protein is then proteolytically cleaved by proprotein convertases (PCs), including furin, PC5/6, paired basic amino acid-cleaving enzyme 4, and PC7.

Signal Transducer and Activator of Transcription 6 (STAT6) is a member of the STAT family of transcription factors. At least seven STATs exist: STAT1, 2, 3, 4, 5a, 5b, and 6. They are responsible for an array of cellular activities including regulating growth, survival, differentiation, motility, and the immune response. STAT6 plays a central role in exerting IL4 mediated biological responses. It is found to induce the expression of BCL2L1/BCL-X(L), which is responsible for the anti-apoptotic activity of IL4. Knockout studies in mice suggested the roles of this gene in differentiation of T helper 2 (Th2) cells, expression of cell surface markers, and class switch of immunoglobulins. STAT6 has been shown to interact with EP300, CREB-binding protein, NFKB1, Nuclear receptor coactivator 1, IRF4 and SND1.

Interleukin 23 receptor (IL23R) is a type I cytokine receptor for IL23. Endothelial cell-specific molecule–1 (ESM-1)—so-called endocan—is a novel endothelium derived soluble dermatan sulphate proteoglycan (PG) that is constitutively expressed by endothelial cells in lungs and kidneys and can be detected in human blood. It is encoded by the ESM1 gene. The expression of this gene is regulated by several cytokines and growth factors, such as vascular endothelial growth factor. Inflammatory cytokines, such as interleukin (IL)-1 beta and tumour necrosis factor (TNF)- alpha, stimulate the upregulation of endocan mRNA and the secretion of endocan from endothelial cells. The binding of circulating endocan to leukocyte ligand for ICAM-1—Lymphocyte Function-associated Antigen-1 (LFA-1) and to leukocyte ligand for VCAM-1—Very Late Antigen-4 (VLA-4) is important in leukocyte adhesion and interaction with activated endothelium. Endocan is a key player in the regulation of major processes such as cell adhesion in inflammatory disorders and tumour progression.
