Heterotrimeric G proteins function to relay information from cell surface receptors to intracellular effectors (1). Each of a very broad range of receptors specifically detects an extracellular stimulus (a photon, pheromone, odorant, hormone or neurotransmitter) while the effectors (i.e., adenylyl cyclase), which act to generate one or more intracellular messengers, are less numerous. In mammals, G protein alpha, Beta and Gamma polypeptides are encoded by at least 16, 4 and 7 genes, respectively (2-5). Most interest in G proteins has been focused on their a subunits, since these proteins bind and hydrolyze GTP and most obviously regulate the activity of the best studied effectors. Four distinct classes of G alpha subunits have been identified; these include Gs, Gi, Gq and Ga 12/13 (3,4). The Gi class comprises all the known a subunits that are susceptible to pertussis toxin modifications, including Ga i-1, Ga i-2, Ga i-3, Ga o, Ga t1, Ga t2, Ga z and Ga gust (4). Of these, the three Ga i subtypes function to open atrial potassium channels (6). Ga 16 is a member of the Gq subfamily and is expressed specifically in hematopoietic cells (7).

Recognizes the L1 or Calprotectin molecule, an intra-cytoplasmic antigen comprising of a 12 kDa alpha chain and a 14 kDa beta chain expressed by granulocytes, monocytes and by tissue macrophages. Macrophages usually arise from hematopoietic stem cells in the bone marrow. Under migration into tissues, the monocytes undergo further differentiation to become multifunctional tissue macrophages. They are classified into normal and inflammatory macrophages. Normal macrophages include macrophages in connective tissue (histiocytes), liver (Kupffer's cells), lung (alveolar macrophages), lymph nodes (free and fixed macrophages), spleen (free and fixed macrophages), bone marrow (fixed macrophages), serous fluids (pleural and peritoneal macrophages), skin (histiocytes, Langerhans's cell) and in other tissues. Inflammatory macrophages are present in various exudates. Macrophages are part of the innate immune system, recognizing, engulfing and destroying many potential pathogens including bacteria, pathogenic protozoa, fungi and helminthes. This MAb reacts with neutrophils, monocytes, macrophages, and squamous mucosal epithelia and has been shown as an important marker for identifying macrophages in tissue sections.

The LRRTM protein family plays a role in the regulation of various cellular events during nervous system development. Localizing predominantly to the nervous system, LRRTM family members are known to exhibit synaptogenic activity. LRRTM3 (leucine rich repeat transmembrane neuronal 3) is a 581 amino acid single-pass type I membrane protein belonging to the LRRTM family. LRRTM3 is involved in the development and maintenance of the vertebrate nervous system, and contains ten LRR repeats. Expressed in neuronal tissues, LRRTM3 is encoded by a gene that maps to a region of chromosome 10 that has been linked to late-onset Alzheimer disease and elevated plasma Beta-Amyloid. As a result of alternative splicing events, two LRRTM3 isoforms exist.

Containing nine LRR repeat and a cleavable signal peptide, the gene encoding LRRC54 maps to human chromosome 11, which houses over 1,400 genes and comprises nearly 4% of the human genome. Jervell and Lange-Nielsen syndrome, Jacobsen syndrome, Niemann-Pick disease, hereditary angioedema and Smith-Lemli-Opitz syndrome are associated with defects in genes that maps to chromosome 11.
