IFI16 is a member of the HIN-200 (hematopoietic interferon-inducible nuclear antigens with 200 amino acid repeats) family of cytokines. The encoded protein contains domains involved in DNA binding, transcriptional regulation, and protein-protein interactions. The protein localises to the nucleoplasm and nucleoli, and interacts with p53 and retinoblastoma-1. It modulates p53 function, and inhibits cell growth in the Ras/Raf signaling pathway. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.

APOBEC2 is a 224 amino acid protein that belongs to the cytidine and deoxycytidylate deaminase family. Expressed exclusively in heart and skeletal muscle, APOBEC2 is thought to be a probable C (cytidine) to U (uridine) editing enzyme. However, unlike other members of the family, such as APOBEC1, which mediates the editing of apolipoprotein (apo) B mRNA, APOBEC2 does not display any detectable apoB mRNA editing activity. Also, APOBEC2 has been shown to have low, but definite, intrinsic cytidine deaminase activity.

FBXO25 is a member of the F box protein family which is characterized by an approximately 40 amino acid motif, the F box. The F box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1 cullin F box), which function in phosphorylation dependent ubiquitination. The F box proteins are divided into 3 classes: Fbws containing WD 40 domains, Fbls containing leucine rich repeats, and Fbxs containing either different protein protein interaction modules or no recognizable motifs. FBXO25 belongs to the Fbxs class. There are three named isoforms produced by alternative splicing.FBXO25, also known as FBX25, is a 367 amino acid protein that contains one C-terminal F-box domain and belongs to the Fbx class of the F-box family of proteins. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and recruitment for ubiquitination. They are members of a larger family of proteins that are involved in the regulation of a wide variety of cellular processes (including the cell cycle, immune response, signaling cascades and developmental processes) through the targeting of proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkB-a and b-catenin, for degradation by the proteasome after ubiquitination. Expressed at high levels in brain, FBXO25 localizes predominantly to the nucleus and directly interacts with Skp1 p19 and CUL-1. Disruption of the gene encoding FBXO25 can lead to X-linked mental retardation.

Bestrophin-4 is a 473 amino acid member of the Bestrophin family of proteins. Members of the Bestrophin family are transmembrane proteins that contain a high percentage of aromatic residues, a conserved RFP (Arg-Phe-Pro) motif and they function as anion channels. Bestrophin-4 acts as a calcium-sensitive chloride channel located in the cell membrane. It is believed that Bestrophin-4 also acts as a channel for other physiologically significant anions, such as bicarbonate. Bestrophin-4 is predominantly expressed in the colon, but can be found at low levels in testis, placenta, trachea, spinal chord, lung and retina.

E2F's are DNA binding proteins, which associate with negative regulators, such as the retinoblastoma p107 protein, resulting in an altered rate of gene transcription. The E2F proteins contain several evolutionally conserved domains found in most members of the family. These domains include a DNA binding domain, a dimerization domain which determines interaction with the differentiation regulated transcription factor proteins (DP), a transactivation domain enriched in acidic amino acids, and a tumor suppressor protein association domain which is embedded within the transactivation domain. This protein and another 2 members, E2F2 and E2F3, have an additional cyclin binding domain. E2F1 is proposed to be involved in several cellular processes that range from tumor suppressor, cell progression and oncogenesis. E2F1 overexpression can also drive cells into apoptosis.
