The fidelity of protein synthesis requires efficient discrimination of amino acid substrates by aminoacyl-tRNA synthetases. Aminoacyl-tRNA synthetases function to catalyze the aminoacylation of tRNAs by their corresponding amino acids, thus linking amino acids with tRNA-contained nucleotide triplets. ProRS (Prolyl-tRNA synthetase), also known as EPRS, EARS, PARS, QARS, QPRS, PIG32 or GLUPRORS, is a 1,512 amino acid protein that contains three WHEP-TRS domains and belongs to both the class-I and class-II aminoacyl-tRNA synthetase family. Functioning as a component of the multisynthase complex, ProRS uses ATP to catalyze the conversion of L-glutamate and tRNA(Glu) to L-glutamyl-tRNA(Glu), as well as the conversion of L-proline and tRNA(Pro) to L-prolyl-tRNA(Pro).

The intronless AHNAK gene is located on human chromosome 11q12 and has three main structural regions: the 251 amino acid N-terminus, a large central region of 4390 amino acids with multiple repeated units of about 128 amino acids in length, and the 1002 amino acid C-terminus. The central region seems to have antiparallel beta-strands connected by intervening loops. Several putative regulatory elements are clustered within the C-terminal region, including nuclear export localization signals, a leucine zipper, and potential phosphorylation sites for Akt1 and PKC. AHNAK is believed to be an important signalling molecule involved in a wide range of physiological activities and may be required for neuronal cell differentiation.. AHNAK also appears to influence b-adrenergic regulation of cardiac L-type Ca2+ channel function.

GPRC6A (G protein-coupled receptor family C group 6 member A) is a 928 amino acid protein encoded by the mouse Gprc6a gene. GPRC6A is a member of the G protein-coupled receptor 3 family. This receptor is activated by both amino acids and extracellular concentration of calcium ions. The activity of GPRC6A is mediated by a G protein that activates a phosphatidylinositol-calcium second messenger system. GPRC6A senses changes in the extracellular concentration of calcium ions, suggesting that it may mediate extracellular calcium-sensing responses in osteoblasts. Osteocalin stimulates the activity of GPRC6A in the presence of calcium. GPRC6A also acts as a receptor for amino acids, with a preference for basic amino acids such as L-Lys, L-Arg and L-ornithine. Its affinity for amino acids suggests that it may act as a regulatory component of the urea cycle. GPRC6A is expressed at high levels in liver, lung, spleen and heart, and at lower levels in kidney, skeletal muscle and brain.
