The leucine-rich (LRR) repeat is a 20 to 30 amino acid motif that forms a hydrophobic å/ horseshoe fold, allowing it to accommodate several leucine residues within a tightly packed core. All LRR repeats contain a variable segment and a highly conserved segment, the latter of which accounts for 11 or 12 residues of the entire LRR motif. SLITRK3 (SLIT and NTRK-like family, member 3) is a 977 amino acid single-pass type I membrane protein that contains 20 LRR repeats and belongs to the SLITRK family. Expressed at highest levels in cerebral cortex, SLITRK3 is also found in adult and fetal neural tissues and some astrocytic brain tumours. SLITRK3 functions to suppress neurite outgrowth and plays a role in the regulation of neuronal function. SLITRK3 is encoded by a gene that maps to human chromosome 3, which houses over 1100 genes, including a chemokine receptor (CKR) gene cluster and a variety of human cancer-related gene loci.

Eukaryotic RNA polymerase II mediates the synthesis of mature and functional messenger RNA. This is a multistep process, called the transcription cycle, that includes five stages: preinitiation, promoter, clearance, elongation and termination. Elongation is thought to be a critical stage for the regulation of gene expression. ELL (11-19 lysine-rich leukemia protein), also designated MEN, functions as an RNA polymerase II elongation factor that increases the rate of transcription by suppressing transient pausing by RNA polymerase II. It is also thought to regulate cellular proliferation. ELL is abundantly expressed in peripheral blood leukocytes, skeletal muscle, placenta and testis, with lower expression in spleen, thymus, heart, brain, lung, kidney, liver and ovary. The gene encoding human ELL, which maps to chromosome 19p13.1, is one of several genes that undergo translocation with the MLL gene on chromosome 11q23 in acute myeloid leukemia. MLL (myeloid/lymphoid leukemia, also designated ALL-1 and HRX) regulates embryonal and hematopoietic development.

The SCA11 gene, comprising of 16 exons, produces a 5.6-kb transcript in which the longest open reading frame is 3,732 nucleotides. Defects in the SCA11 protein are the cause of the disorder spinocerebellar ataxia type 11 (SCA11). Spinocerebellar ataxia is a clinically and genetically heterogeneous group of cerebellar disorders. Patients show progressive incoordination of gait and often poor coordination of hands, speech and eye movements, due to degeneration of the cerebellum with variable involvement of the brainstem and spinal cord. The SCA11 disorder is an autosomal dominant cerebellar ataxia (ADCA). It is a relatively benign, late-onset, slowly progressive neurologic disorder. The SCA11 protein has also been implicated in Alzheimer disease and in tangle formation. Existing as three alternatively spliced isoforms, the SCA11 gene is conserved in chimpanzee, dog, cow, mouse, rat, chicken and zebrafish, and maps to human chromosome 15q15.2.

The tripartite motif (TRIM) family of proteins are characterized by a conserved TRIM domain that includes a coiled-coil region, a B-box type zinc finger, one RING finger and three zinc-binding domains. Staf-50 (50 kDa-stimulated trans-acting factor), also known as TRIM22 (tripartite motif-containing 22), RNF94 or GPSTAF50, is a 498 amino acid cytoplasmic protein that belongs to the TRIM family and, characteristic of TRIM family members, contains one RING-type zinc finger, one B box-type zinc finger and one SPRY domain. Induced by IFN-å and IFN-∫, Staf-50 is strongly expressed in ovary, spleen, thymus and peripheral blood leukocytes where it is thought to mediate the antiviral effects of IFN proteins. Additionally, Staf-50 is present in leukemic cells, suggesting a role in cancer formation and metastasis. Staf-50 exists as two alternatively spliced isoforms which are encoded by a gene that maps to human chromosome 11.

Cadherin-11 is a type II classical cadherin member of the cadherin superfamily of integral membrane proteins that mediate calcium-dependent cell-cell adhesion. Cadherins interact with themselves in a homophilic manner in connecting cells, and thus contribute to the sorting of heterogeneous cell types. Cadherin-11 contains five cadherin domains and is mainly expressed in the brain. Mature cadherin proteins consists of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small highly conserved C-terminal cytoplasmic domain. It is shown that Cadherin-11 is a viable molecular target for therapeutic intervention in Glioblastoma multiforme.
