Muskelin is a 735 amino acid nuclear and cytoplasmic protein that mediates cytoskeletal responses and cell spreading to Thrombospondin 1, an extracellular matrix glycoprotein. Forming a CTLH complex with RMND5A, Ran BP-M and TWA1, Muskelin exists as two alternatively spliced isoforms. Muskelin is implicated as an isoform-specific anchoring protein for the prostaglandin EP3 receptor, and contains one LisH domain, a CTLH domain, six Kelch repeats and an N-terminal discoidin-like domain through which it self-associates via a head-to-tail mechanism. Human and mouse Muskelin share 98% amino acid seqeuence homology and are encoded by genes located on chromosomes 7q32.3 and 6 A3.3, respectively.

Excitatory Amino Acid Transporters (EAATs) are membrane-bound proteins that are localized in glial cells and pre-synaptic glutamatergic nerve endings. EAATs transport the excitatory neurotransmitters L-glutamate and D-aspartate, a process that is essential for terminating the postsynaptic action of glutamate. The re-uptake of amino acid neurotransmitters by EAAT proteins has been shown to protect neurons from excitotoxicity, which is caused by the accumulation of amino acid neurotransmitters. EAAT4 is an aspartate/glutamate transporter that is expressed predominantly in the cerebellum. The transport activity encoded by EAAT4 has high apparent affinity for L-aspartate and L-glutamate, and has a pharmacologic profile consistent with previously described cerebellar transport activities. EAAT5 is a glutamate transporter coupled to a chloride conductance which is expressed primarily in retina. Although EAAT5 shares the structural homologies of the EAAT family, a novel feature of the EAAT5 sequence is a carboxy-terminal motif previously identified in N-ethyl-D-aspartate receptors and potassium channels and shown to confer interactions with a family of synaptic proteins that promote ion channel clustering.

Pbx 1, 2, 3 and 4 are members of the TALE (three amino acid loop extension) family of homeodomain-containing proteins. Human pre-B cell acute leukemias are frequently associated with a t(1;19)(q23;p13.3) chromosomal rearrangement, which creates a chimeric gene encoding a fusion between the E2A and Pbx 1 gene products. Pbx 2 and Pbx 3 share 92% and 94% respective identities with Pbx 1 over a 266 amino acid region flanking their homeobox domains, while all three proteins are quite divergent at their amino- and carboxy-termini. Two forms of Pbx 1 and Pbx 3 each differ primarily in their carboxy-termini and result from alternative mRNA splicing. Unlike other homeotic selector genes which are expressed transiently during development and differentiation, Pbx gene transcripts are ubiquitously expressed in both fetal and adult tissues and cell lines. Additionally, Pbx 2 and Pbx 3 transcripts are detected in lymphoid cells, which do not express Pbx 1. Pbx 4 expression is confined to the testis, especially to spermatocytes in the pachytene stage of the first meiotic prophase.
