HDAC7 is a member of the class II mammalian histone deacetylases, which plays an important role in modulating the eukaryotic chromatin structure. Human HDAC7 is composed of 912 amino acid residues. Although HDAC7 is localized mostly to the cell nucleus, it is also found in the cytoplasm, suggesting nucleo-cytoplasmic shuttling. The histone deacetylase activity of HDAC7 maps to a carboxy-terminal domain and is dependent on interaction with class I HDACs in the nucleus. It is an active component of different transcriptional corepressor complexes that can be recruited to specific promoter regions via interactions with a growing number of sequence specific transcriptional factors. HDAC7 catalyzes removal of acetyl-groups from acetyl-lysines of histones and promotes compaction of chromatin in these regions, leading to the inhibition of gene transcription.

Glut4 is a twelve pass transmembrane protein (12TM) whose carboxy-terminus may dictate its cellular localization. Aberrant Glut4 expression has been suggested to contribute to such maladies as obesity and diabetes. Glut4 null mice have shown that while functional Glut4 protein is not required for maintaining normal glucose levels, it is necessary for sustained growth, normal cellular glucose, fat metabolism and prolonged longevity. TUG (ASPL in humans) regulates the trafficking of glucose via Glut4. Full-length TUG forms a complex with Glut4 and in 3T3-L1 adipocytes and this complex is present in unstimulated cells and is disassembled by insulin. TUG acts by trapping endocytosed Glut4 and tethering it intracellularly. Insulin mobilizes this pool of retained Glut4 by releasing the tether.

The large chondroitin sulfate proteoglycan, aggrecan, is the predominant proteoglycan present in cartilage. Aggrecan is a member of the chondroitin sulphate proteoglycan family, which also includes versican/PG-M, neurocan and brevican. Aggrecan is a complex multidomain macromolecule that undergoes extensive processing and post-translational modification. Aggrecan in cartilage forms aggregates with hyaluronan and link protein, embedded in a collagen network. Aggrecan accounts for the compressive stiffness and resilience of the hyaline cartilage. Many forms of inflammatory arthritis are shown to be accompanied with aggrecan degradation and loss from the cartilage. Brevican is a brain proteoglycan of the aggrecan/versican/neurocan family. In the adult brain, the brevican core protein undergoes proteolytic cleavage and exists as a full-length form a carboxy-terminal fragment and an amino-terminal fragment.

HP1 proteins are relatively small proteins (~25 kDa) with a conserved amino-terminal chromo domain and a structurally related carboxy-terminal motif, the chromo shadow domain. Both domains of HP1 are required for binding to native chromatin in vivo, but they contribute differentially to binding in euchromatin and heterochromatin.

The P2X receptor family is comprised of ligand-gated ion channels that allow for the increased permeability of calcium into the cell in response to extracellular ATP. The seven P2X receptors, P2X1-P2X7, form either homomeric or heteromeric channels or both. They are characterised by intracellular amino- and carboxy-termini. P2X receptors are expressed in a wide variety of tissues, including neurons, prostate, bladder, pancreas, colon, testis and ovary. The major function of the P2X receptors is to mediate synaptic transmissions between neurons and to other tissues via the binding of extracellular ATP, which acts as a neurotransmitter. The P2X receptors may be involved in the onset of necrosis or apoptosis after prolonged exposure to high concentrations of extracellular ATP.
