Schlafen proteins function as suppressors of cell growth and are thought to play a role in the maintenance of T cell quiescence. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Cytochrome c oxidase, the terminal component of the mitochondrial respiratory chain, catalyzes the electron transfer from reduced cytochrome c to oxygen. This component is a heteromeric complex consisting of three catalytic subunits encoded by mitochondrial genes, and multiple structural subunits encoded by nuclear genes. The mitochondrially-encoded subunits function in electron transfer, while the nuclear-encoded subunits may function in the regulation and assembly of the complex. This nuclear gene encodes polypeptide 2 (liver isoform) of subunit VIIa, with this polypeptide being present in both muscle and non-muscle tissues. In addition to polypeptide 2, subunit VIIa includes polypeptide 1 (muscle isoform), which is present only in muscle tissues, and a related protein, which is present in all tissues. Alternative splicing results in multiple transcript variants. Related pseudogenes have been identified on chromosomes 4 and 14.

Bad is a member of the Bcl2 family and acts to promote apoptosis by forming heterodimers with the survival proteins Bcl2 and BclxL, thus preventing them from binding with BAX. Bad is found on the outer mitochondrial membrane and, once phosphorylated in response to growth stimuli, translocates to the cytoplasm. The phosphorylation status of Bad represents a key checkpoint for death or cell survival. JNK-induced phosphorylation of BAD serine 128 promotes the apoptotic role of Bad by opposing the inhibitory effect of growth factor on Bad-mediated apoptosis. Cdc2-induced phosphorylation of Bad serine 128 has an inhibitory effect on its interaction with 14-3-3 proteins. The latter interaction is critical for Bad phosphorylation at serine 155, a site within the BH3 domain that leads to the release of BclxL and the promotion of cell survival. Alternative splicing of this gene results in two transcript variants which encode the same isoform.

The sterile alpha motif (SAM) domain is a 70 residue structure found in a large number of proteins involved in diverse processes present throughout eukaryotes. The SAM domain is known to bind RNA and is arranged in a small five-helix bundle with two large interfaces. SAMD14 (sterile alpha motif domain-containing protein 14) is a 417 amino acid protein encoded by the SAMD14 gene which maps to human chromosome 17. Chromosome 17 makes up over 2.5% of the human genome with about 81 million bases encoding over 1,200 genes. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Chromosome 17 is also linked to neurofibromatosis, a condition characterized by neural and epidermal lesions, and dysregulated Schwann cell growth, Alexander disease, Birt-Hogg-Dube syndrome and Canavan disease.

Cytokeratin 14 (CK14) belongs to the type I (or A or acidic) subfamily of low molecular weight keratins and exists in combination with keratin 5 (type II or B or basic). CK14 is found in basal cells of squamous epithelia, some glandular epithelia, myoepithelium, and mesothelial cells. Anti-CK14 is useful in differentiating squamous cell carcinomas from poorly differentiated epithelial tumors. Anti-CK14 is one of the specific basal markers for distinguishing between basal and non-basal subtypes of breast carcinomas. Anti-CK14 is also a good marker for differentiation of intraductal from invasive salivary duct carcinoma by the positive staining of basal cells surrounding the in-situ neoplasm as well as for differentiation of benign prostate from prostate carcinoma. Furthermore, this antibody has been useful in separating oncocytic tumors of the kidney from its renal mimics, and in identifying metaplastic carcinomas of the breast.

Recognizes a protein of14 kDa, identified as MRP-14 (also known as Calgranulin B or S100AA9). It comprises 60% of the cytoplasmic protein fraction of circulating polymorphonuclear granulocytes and is also found in monocytes, macrophages and ileal tissue eosinophils. Peripheral blood monocytes carry the antigen extra- and intracellularly, neutrophils only intracellularly. It is a potent chemotactic factor for neutrophils. Plasma concentrations are elevated in diseases associated with increased neutrophil activity, like inflammatory bowel disease. Granulocytes terminate their existence after transmigration through the intestinal wall. Therefore, it is also detectable in feces. Elevated levels have been observed in body fluids such as plasma, saliva, gingival crevicular fluid, stools, and synovial fluid during infection and inflammatory conditions. This MAb reacts with neutrophils, monocytes, and macrophages, and has been shown as an important marker for identifying macrophages in tissue sections.
