SNAP 29 is a 258 amino acid protein that localizes to the membrane and the cytoplasm, as well as to the cell junction, and contains one t-SNARE coiled-coil homology domain. Expressed in liver, heart, brain, kidney, placenta, lung, spleen, pancreas and skeletal muscle, SNAP 29 binds tightly to Syntaxins and, via this binding, is involved in membrane trafficking events. Defects in the gene encoding SNAP 29 are the cause of CEDNIK syndrome, a neurocutaneous syndrome that is associated with cerebral dysgenesis, neuropathy, ichthyosis and palmoplantar keratoderma. The gene encoding SNAP 29 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Kainate/AMPA receptors are co-localized with NMDA receptors in many synapses and consist of seven structurally related subunits designated GluR-1 to -7. The kainate/AMPA receptors are primarily responsible for the fast excitatory neuro-transmission by glutamate, whereas the NMDA receptors are functionally characterized by a slow kinetic and a high permeability for Ca2+ ions. The NMDA receptors consist of five subunits: epsilion 1, 2, 3, 4 and one zeta subunit. The zeta subunit is expressed throughout the brainstem, whereas the four epsilon subunits display limited distribution.

As a subfamily of the cadherin superfamily, protocadherins are cadherin-like cell adhesion proteins that contain up to seven extracellular domains and are predominantly expressed in the nervous system. Importantly, the adhesion mechanism of protocadherins is distinct from classic cadherins. Through inactivation or overexpression, several protocadherins have been implicated in a variety of cancers. Protocadherin-20 (PCDH20), also known as protocadherin-13, is a 924 amino acid protein containing 6 cadherin domains and potentially functioning as a calcium-dependent cell-adhesion protein. In non-small cell lung cancer cell lines, a homozygous loss of PCDH20 was identified through either deletion of one allele and methylation of the other or methylation of both alleles. Hypermethylation of PCDH20 is associated with worse prognosis and clinical outcome, suggesting that PCDH20 may function as a tumor suppressor.

The X and Y chromosomes are the human sex chromosomes. Chromosome X consists of about 153 million base pairs and nearly 1000 genes. The combination of an X and Y chromosome lead to normal male development while two copies of X lead to normal female development. There are a number of conditions related to an unusual number and combination of sex chromosomes being inherited. More than one copy of the X chromosome with a Y chromosome causes Klinefelter's syndrome. A single copy of X alone leads to Turner's syndrome. More than 2 copies of the X chromosome, in the absence of a Y chromosome, is known as Triple X syndrome. Color blindness, hemophilia, and Duchenne muscular dystrophy are well known X chromosome-linked conditions which affect males more frequently as males carry a single X chromosome. The CXX1 gene product has been provisionally designated CXX1 pending further characterisation.

TorsinB is a 336 amino acid protein that localizes to the lumen of endoplasmic reticulum and belongs to the Torsin family. Expressed in a variety of tissues with particularly high levels in adult brain, TorsinB is thought to function as a molecular chaperone that assists in the conformational folding of membrane and secreted proteins. Via its ability to regulate protein folding, TorsinB plays a key role in postnatal developmental events and is essential for proper neurological development. TorsinB contains two PKC (protein kinase C) phosphorylations sites, one ATP binding domain and several conserved cysteines and shares 69% amino acid identity with TorsinA, a related family member. Defects in the gene encoding TorsinB are associated with torsion dystonia, an autosomal dominant neurological disorder that is characterized by debilitating muscle contractions throughout the body.

GTP-binding protein 9 is a 396 amino acid protein that belongs to the Obg-related GTPase family under the translation factors (TRAFAC) class. Originally thought to only have GTPase activity, Obg-related GTPase family members have been shown to also have ATPase activity. In Homo sapians, GTPBP9 exhibits a preference for binding ATP over GTP, with GTP binding occuring only at high nucleotide concentration. One cause for ATP affinity and GTP discrimination is thought to be a substitution of glutamine for a hydrophobic amino acid in Obg-related family members; this is the same substitution that inactivates Ras-like GTPases. GTPBP9 contains a C-terminal TGS domain that binds to ligands and an N-terminal G domain which binds nucleotides. GTPBP9 is expressed as three isoforms produced by alternative splicing.

Spliceosomes are multi-protein complexes that are composed of snRNPs (small nuclear ribonucleoproteins) and a variety of associated protein factors, all of which work in concert to regulate the splicing of pre-mRNA, a critical step in the post-transcriptional regulation of gene expression. CWC22 (CWC22 spliceosome-associated protein), also known as NCM, fSAPb or EIF4GL, is a 908 amino acid nuclear protein and component of the spliceosome C complex. CWC22 is associated with the spliceosome prior to catalytic steps and remains associated throughout the reaction. Belonging to the CWC22 family, CW22 contains one MI domain and a MIF4G domain. The gene encoding CWC22 maps to human chromosome 2q31.3. As the second largest human chromosome, chromosome 2 makes up approximately 8% of the human genome and contains 237 million bases encoding over 1,400 genes.

A crucial regulator of chromatin dynamics and DNA transcription is the covalent modification and methylation of histones. Generally, methylation of certain lysine residues on Histone H3 and Histone H4 can be associated with transcriptionally active or inactive chromatin. This regulation has profound effects on the expression of genes and is part of an epigenetic memory network that determines cell fate. JMJD7 (Jumonji domain-containing protein 7) is a member of a family of JMJC domain-containing histone demethylases that are directly involved in removing methyl residues from distinct and unique lysine residues. These actions are implicated in gene expression and the regulation of cell senescence. JMJC domain-containing histone demethylases are also likely involved in development via their ability to regulate gene expression. JMJD7 contains one JMJC domain and was originally thought to be an alternatively spliced isoform of PLA2G4B.

PIST (PDZ protein interacting specifically with TC10), also known as GOPC (golgi associated PDZ and coiled-coil motif containing), CAL or FIG, is a 462 amino acid protein that localizes to the cytoplasm, as well as to the membrane of the golgi apparatus and to the cell junction. Expressed ubiquitously and containing one PDZ (DHR) domain, PIST functions as a homooligomer that interacts with a variety of proteins and plays a role in intracellular protein trafficking and degradation. Additionally, PIST is thought to regulate ionic currents via membrane channel modification and may also play a role in autophagy. Chromosomal aberrations in the gene encoding PIST are found in glioblastoma multiform (GBM), a common and aggressive form of brain tumour, suggesting a role for mutated PIST in carcinogenesis. Three isoforms of PIST exist due to alternative splicing events.

Fe65L is a 758 amino acid protein that contains one WW domain and two PID domains. Binding to the intracellular domain of the ∫-Amyloid precursor protein, Fe65L is thought to modulate the internalization and, therefore, the accessibility and function of ∫-Amyloid. Via its ability to control the intracellular accumulation of ∫-Amyloid, Fe65L is thought to play a role in the pathogenesis of Alzheimer's disease. Multiple isoforms of Fe65L exist due to alternative splicing events. The gene encoding Fe65L maps to human chromosome 4, which encodes nearly 6% of the human genome and has the largest gene deserts (regions of the genome with no protein encoding genes) of all of the human chromosomes. Defects in some of the genes located on chromosome 4 are associated with Huntington's disease, Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease.

Sorting nexin (SNX) proteins are members of a large family of hydrophilic PX (phospholipid-binding motif) domain-containing proteins that interact with a variety of receptor types. SNXs are widely expressed, although the tissue distribution of each SNX mRNA varies. The ability of SNXs to bind specific phospholipids, as well as their tendency to form protein-protein complexes, suggests a role for these proteins in cellular membrane trafficking and protein sorting. SNXs may also function specifically in pro-degradative sorting, internalization, endosomal recycling or simply in endosomal sorting. SNX10 (Sorting nexin-10) is a 201 amino acid protein that contains one phox domain and belongs to the SNX family. Like other members of the SNX family, SNX10 is thought to play a role in intracellular trafficking events throughout the cell.

The DnaJ family is one of the largest of all the chaperone families and has evolved with diverse cellular localization and functions. The presence of the J domain defines a protein as a member of the DnaJ family. DnaJ heat shock induced proteins are from the bacterium Escherichia coli and are under the control of the htpR regulatory protein. The DnaJ proteins play a critical role in the HSP 70 chaperone machine by interacting with HSP 70 to stimulate ATP hydrolysis. The proteins contain cysteine rich regions that are composed of zinc fingers that form a peptide-binding domain responsible for the chaperone function. DnaJ proteins are important mediators of proteolysis and are in-volved in the regulation of protein degradation, exocytsis and endocytosis. DnaJC7 (DnaJ homolog subfamily C member 7), also known as TPR2, TTC2 or DANJC7, is ubiquitously expressed, with highest expression in testis, liver, heart and brain.
