Beschreibung:   The 26S proteasome is a large complex involved in the intracellular degradation of proteins in eukaryotes. Ubiquitination by E3 ubiquitin ligases targets proteins for degradation by this complex. The 26S proteasome plays an important role in the regulation of many biological processes. It is composed of over 30 different subunits and contains at least two copies of each subunit. Assembly of this large complex is ATP-dependent. Due to its size, it is fairly unstable and often disassociates into subcomplexes (including a 20S core and two 19S regulatory complexes). The 26s proteasome p42A (also known as Rpn7 in yeast and S10 in human) is one of at least nine, non-ATPase lid subunits of the 19S regulatory complex. It is important in the proper assembly and stability of the 26S proteasome. The 19S regulatory complex recognizes ubiquitinated proteins, removes the ubiquitin chains and translocates the proteins to the 20S core for degradation.

Beschreibung:   The ubiquitin (Ub) pathway involves three sequential enzymatic steps that facilitate the conjugation of Ub and Ub-like molecules to specific protein substrates. The first step requires the ATP-dependent activation of the Ub C-terminus and the assembly of multi-Ub chains by the Ub-activating enzyme known as the E1 component. The Ub chain is then conjugated to the Ub-conjugating enzyme (E2) to generate an intermediate Ub-E2 complex. The Ub-ligase (E3) then catalyzes the transfer of Ub from E2 to the appropriate protein substrate. UBE2E1 and UBE2L3, also designated UBCH6 and UBCH7 respectively in human, are E2 conjugating enzymes that interact with various proteins. Specifically, UBE2E1 interacts with the tumor suppressor protein TSSC5. UBE2L3 has been shown to mediate c-fos degradation, NF-kB maturation, human papilloma virus-mediated p53 and Myc protein degradation.

Beschreibung:   FBXO29, also designated F-box/WD repeat-containing protein 8 (FBXW8), is a 598 amino acid protein that contains one 40 amino acid F-box region, making it a member of the F-box family. FBXO29 also contains five WD repeats. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein) type E3 ubiquitin ligase complex and are involved in substrate recognition and recruitment for ubiquitination. F-box proteins are members of a large family that regulates cell cycle, immune response, signaling cascades and developmental programs by targeting proteins, such as cyclins, cyclin-dependent kinase inhibitors, IkB-å and -catenin, for degradation by the proteasome after ubiquitination. Functioning as a component of the SCF complex, FBXO29 is thought to recognise and bind to select phosphorylated proteins, thereby promoting their ubiquitination and subsequent degradation. FBXO29 exists as two isoforms as a result of alternative splicing events.

Beschreibung:   The protein encoded by this gene belongs to the SMAD, a family of proteins similar to the gene products of the Drosophila gene 'mothers against decapentaplegic' (Mad) and the C. elegans gene Sma. SMAD proteins are signal transducers and transcriptional modulators that mediate multiple signaling pathways. This protein mediates the signals of the bone morphogenetic proteins (BMPs), which are involved in a range of biological activities including cell growth, apoptosis, morphogenesis, development and immune responses. In response to BMP ligands, this protein can be phosphorylated and activated by the BMP receptor kinase. The phosphorylated form of this protein forms a complex with SMAD4, which is important for its function in the transcription regulation. This protein is a target for SMAD-specific E3 ubiquitin ligases, such as SMURF1 and SMURF2, and undergoes ubiquitination and proteasome-mediated degradation. Alternatively spliced transcript variants encoding the same protein have been observed. [provided by RefSeq].

Beschreibung:   SH3MD2 acts as a scaffold protein, contributes to Rac-induced signal transduction such as JNKs (MAPK8 and MAPK9) activation and induces apoptosis. Within a signaling complex, it probably recruits protein kinases such as MAP3K10 or MAP3K11 which are in turn activated leading to the sequential activation of MAP2K4, MAP2K7 and JNKs (MAPK8 and MAPK9). SH3MD2 may be involved in targeting of HIV-1 GAG and GAG-POL polyproteins to the plasma membrane. This gene encodes a protein containing an N-terminus RING-finger, four SH3 domains, and a region implicated in binding of the Rho GTPase Rac. Via the RING-finger, the encoded protein has been shown to function as an ubiquitin-protein ligase involved in protein sorting at the trans-Golgi network. The encoded protein may also act as a scaffold for the c-Jun N-terminal kinase signaling pathway, facilitating the formation of a functional signaling module. There are two named isoforms.

Beschreibung:   Tripartite motif-containing protein 32 (TRIM32) belongs to the tripartite motif (TRIM) protein family. TRIM32, like all TRIM proteins, contains a domain structure composed of a B-box, a RING-finger and a coiled-coil motif. Additionally, TRIM32 has six C-terminal NHL domains; it is expressed mainly in the skeletal muscle. The TRIM32 gene encodes an E3 ubiquitin ligase, a protein that attaches ubiquitin to a lysine residue on a target protein and acts in conjunction with ubiquitin-conjugating enzymes UbcH5a, UbcH5c and UbcH6. Mutations in the TRIM32 gene cause two forms of autosomal recessive muscular dystrophy designated limb girdle muscular dystrophy type 2H (LGMD2H) and sarcotubular myopathy (STM). TRIM32 mutations can also result in Bardet-Biedl syndrome (BBS), an autosomal recessive disorder characterized by pigmentary retinopathy, polydactyly, hypogenitalism, renal abnormalities, learning disabilities and obesity.

Beschreibung:   CBLL1, also known as HAKAI (meaning ‘destruction’ in Japanese), or RNF188 (RING finger protein 188), is a 491 amino acid protein that contains one C2H2-type zinc finger and one RING-type zinc finger. CBLL1 is believed to function as an E3 ubiquitin-protein ligase that accepts a ubiquitin residue from an E2 ubiquitin-conjugating enzyme and immediately transfers that residue to a protein that is targeted for degradation. More specifically, upon activation of c-Src, CBLL1 interacts with and ubiquitinates tyrosine-phosphorylated E-cadherin, thereby targeting the E-cadherin complex for endocytosis and disrupting epithelial cell-cell contacts. Via its role as an E-cadherin regulator, CBLL1 participates in cell adhesion and may also be involved in the regulation of epithelial-mesenchymal transitions.

Beschreibung:   Ubiquitination is an important molecular mechanism by which abnormal or short-lived proteins are targeted for degradation by the concerted efforts of at least three classes of enzymes: ubiquitin-activating enzymes (E1s), ubiquitin-conjugating enzymes (E2s) and ubiquitin-protein ligases (E3s). UBE2J2 (Ubiquitin-conjugating enzyme E2 J2), also known as NCUBE2 (Non-canonical ubiquitin-conjugating enzyme 2), is a 259 amino acid single pass type IV membrane protein that that belongs to the E2 ubiquitin-conjugating enzyme family and is involved in protein degradation. Localized to the membrane of the endoplasmic reticulum (ER), UBE2J2 catalyzes the attachment of ubiquitin to misfolded membrane proteins, thereby targeting them for proteasomal destruction. This ATP-dependent reaction yields AMP, a diphosphate and a ubiquitin-tagged protein and may be a method of quality control within the ER. Two isoforms of UBE2J2 exist due to alternative splicing events.
