GPR110 (G protein-coupled receptor 110), also known as PGR19, is a 911 amino acid protein that belongs to the G-protein coupled receptor 2 family and LN-TM7 subfamily. Characterized as an adhesion GPCR, GPR110 is a multipass membrane-bound protein with a long amino-terminus that contains multiple domains. One of these domains is the GPCR proteolytic site (GPS), which is essential for proteolytic cleavage of the amino-terminus and for cell surface expression.

SUMF1 is a 374 amino acid alternatively spliced protein that localizes to the lumen of the endoplasmic reticulum and belongs to the sulfatase-modifying factor family. Expressed ubiquitously with highest expression in liver, kidney and pancreas, SUMF1 exists as either a monomer, a homodimer or a heterodimer (with SUMF2) and functions to oxidize sulfatase cysteine residues to an active FGIy residue, thereby playing an important role in sulfatase activity. Defects in the gene encoding SUMF1 are the cause of multiple sulfatase deficiency (MSD), a heterogeneous disorder characterized by metachromatic leukodystrophy, mucopolysaccharidosis, chondrodysplasia punctata, hydrocephalus, ichthyosis, neurologic deterioration and developmental delay.

A translocation between chromosomes 5 and 12, that fuses this gene to that of the translocation, ETV6, leukemia gene, results in chronic myeloproliferative disorder with eosinophilia. Catalytic component of the PR-DUB complex, a complex that specifically mediates deubiquitination of histone H2A monoubiquitinated at 'Lys-119' (H2AK119ub1). Does not deubiquitinate monoubiquitinated histone H2B. Acts as a regulator of cell growth by mediating deubiquitination of HCFC1 N-terminal and C-terminal chains, with some specificity toward 'Lys-48'-linked polyubiquitin chains compared to 'Lys-63'-linked polyubiquitin chains. Deubiquitination of HCFC1 does not lead to increase stability of HCFC1. Interferes with the BRCA1 and BARD1 heterodimer activity by inhibiting their ability to mediate ubiquitination and autoubiquitination. It however does not mediate deubiquitination of BRCA1 and BARD1. Able to mediate autodeubiquitination via intramolecular interactions to couteract monoubiquitination at the nuclear localization signal (NLS), thereby protecting it from cytoplasmic sequestration (PubMed:24703950). Acts as a tumor suppressor.

Junctophilins are components of the junctional complexes between plasma membranes and endoplasmic or sarcoplasmic reticulums present in all excitable cells. Junctophilins contain a cytoplasmic domain which binds to the plasma membrane, as well as an ER/SR membrane spanning hydrophobic C-terminal segment. The three subtypes in this family are Junctophilin-1, -2 and -3. Junctophilin-1 is predominantly expressed in skeletal muscle, but is also expressed at low levels in heart. Junctophilin-2 is expressed in heart and skeletal muscle. Mutant mice lacking the Jph2 gene exhibit embryonic lethality and possess cardiac myocytes that express abnormal calcium transients. Junctophilin-3 is expressed in brain. The JPH3 alternatively spliced exon 2A has been suggested as a site for CTG repeat expansion leading to a Huntington disease-like autosomal dominant disorder.

The Eph subfamily represents the largest group of receptor protein tyrosine kinases identified to date (1–3). While the biological activities of these receptors have yet to be determined, there is increasing evidence that they are involved in central nervous system function and in development (1–3). The Eph subfamily receptors of human origin (and their murine/avian homologs) include EphA1 (Eph), EphA2 (Eck), EphA3 (Hek4), EphA4 (Hek8), EphA5 (Hek7), EphA6 (Hek12), EphA7 (Hek11/MDK1), EphA8 (Hek3), EphB1 (Hek6), EphB2 (Hek5), EphB3 (Cek10, Hek2), EphB4 (Htk), EphB5 (Hek9) and EphB6 (Mep). Ligands for Eph receptors include ephrin-A4 (LERK-4) which binds EphA3 and EphB1. In addition, ephrin-A2 (ELF-1) has been described as the ligand for EphA4, ephrin-A3 (Ehk1-L) as the ligand for EphA5 and ephrin-B2 (Htk-L) as the ligand for EphB4 (Htk) (4–7).

The Ovo family of zinc-finger transcription factors encode evolutionarily conserved genes including those from Caenorhabditis elegans, Drosophila melanogaster, mouse and human. Members of the Ovo family include Ovol1 and Ovol2. Ovol1 acts as a transcriptional repressor by interacting with key developmental signaling pathways such as Wnt and TGF-∫/BMP. Specifically, Ovol1 represses c-Myc and Id2 genes and establishes a balance between proliferation and differentiation of progenitor cells. Deletion of Ovol1 in mice leads to germ cell degeneration and defective sperm production in adult males. Ovol1 has also been shown to repress itself as well as Ovol2, which is thought to regulate neural development and vascular angiogenesis during embryogenesis.

Cyclin Y is a 341 amino acid protein belonging to the cyclin family. Cyclin Y exists as three alternatively spliced isoforms and contains a cyclin N-terminal domain. Cyclin Y may control cell division cycles and regulate cyclin-dependent kinases.Cell proliferation is controlled at specific stages of the cell cycle by distinct protein kinase complexes. These complexes consist of a catalytic subunit associating with a specific regulatory subunit to form the active kinase. The cyclins, which include cyclin A, B, C, D, E, F, G, H, I, K, L, T, Y and their related proteins, including Dbf4, comprise the regulatory subunits of these kinase complexes. The controlled activation of the kinase complexes at various intervals of the cell cycle is regulated by the availability of the cyclins to the catalytic subunit. Unlike the catalytic subunit, which is expressed continually, the expression and stability of the regulatory subunit fluctuates depending on the stage of the cell cycle, thereby regulating kinase activity.
