PIBF is synthesized during pregnancy in response to progesterone by progesterone receptor-positive T lymphocytes (mostly gamma-delta T cells). In the presence of PIBF, natural killer (NK) cells inhibit the release of perforin from storage granules and therefore fail to lyse target cells. In humans, the amount of cells that express PIBF is significantly higher in healthy pregnant women than in women at risk for premature pregnancy termination. Full-length PIBF is associated with the nucleus, whereas secretion of shorter forms is induced by activation of the cell. Research suggests that PIBF functions as a transcription factor in its full-length form, while smaller forms may act as cytokines. The PIBF gene encodes a deduced hydrophilic 757-amino acid alpha-helical protein with an N-terminal signal sequence, a leucine zipper motif, a basic zipper sequence, a PEST sequence, a nuclear localization signal, an endoplasmic reticulum membrane retention signal, and many presumeed N-glycosylation and phosphorylation sites.

Arginase I which is expressed almost exclusively in the liver, catalyzes the conversion of arginine to ornithine and urea . The human arginase I gene, which maps to chromosome 6q23, encodes a 322 amino acid protein. Arginase I exists as a homotrimeric protein and contains a binuclear manganese cluster. Arginase II catalyzes the same reaction as arginase I, but differs in its tissue specificity and subcellular location. Specifically, arginase II localizes to the mitochondria. Arginase II is expressed in non-hepatic tissues, with the highest levels of expression in the kidneys, but, unlike arginase I, is not expressed in liver. The human arginase II gene, which maps to chromosome 14q24.1-q24.3, encodes a 354 amino acid protein. In addition, arginase II contains a putative amino-terminal mitochondrial localization sequence.

ERMAP is a single-pass type one membrane protein that belongs to the immunoglobulin superfamily. Expressed in cord blood, fetal liver and adult bone marrow, ERMAP is thought to function as a cell adhesion molecule in erythroid cells and is responsible for expression of the Scianna/Radin (Sc/Rd) blood group antigen system. The Sc/Rd system is comprised of seven antigens that are present on the surface of red blood cells and have a variety of functions ranging from protein transport to cell adhesion. These seven blood antigens can differ in their expression within a population and may sometimes differ between mother and child. A fetus expressing different blood antigens than its mother may cause the mother to produce against the fetal blood. This condition is known as hemolytic disease of the newborn (HDN) and is characterized by jaundice, anemia and in some cases, infant death.

IRP-1 and IRP-2 are important soluble regulatory factors that mediate iron uptake and storage in mammalian cells. IRPs respond to stress mediators, iron concentration and signaling factors, including nitrogen monoxide, cytokines and hydrogen peroxide.

Serotonin (also designated 5-hydroxytryptamine or 5-HT) is a molecule that functions as a neurotransmitter, a hormone and a mitogen, and it is predominantly expressed in the gut, platelets and central nervous system (CNS). In the CNS, serotonin modulates several processes, including anxiety, sleep, appetite, behavior and drug abuse. In platelets and gut, serotonin plays a major role in cardiovascular function and motility of the gastrointestinal tract, respectively. Serotonin mediates its effects through several of G protein coupled receptors, designated 5-HT receptors or alternatively SR receptors. The SR-2 receptors are comprised of three subtypes, SR-2A, SR-2B and SR-2C, which activate phospholipase C and release intracellular stores of calcium in response to serotonin. SR-2A has a specific role in tracheal smooth muscle contraction, bronchoconstriction and mediating aldosterone production, and it is also thought to play a role in several psychiatric disorders, including depression and schizophrenia. SR-2B is expressed in embryonic and adult cardiovascular tissues, gut and brain and plays an important role in the pathology of cardiac disorders. SR-2C is thought to mediate the effects of atypical antipsychotic drugs.

ARFRP1 (ADP-ribosylation factor related protein 1), also known as ARP or ARL18, is a 201 amino acid membrane-associated GTPase that localizes to the plasma membrane and the Golgi apparatus and is related to the ARF family of regulatory proteins. Expressed in a variety of tissues, ARFRP1 interacts with SYS1 and is thought to be involved in plasma membrane-related signaling events. ARFRP1 exists as multiple alternatively spliced isoforms and is encoded by a gene which maps to a gene cluster on chromosome 20 that is commonly overexpressed in tumors, suggesting a role for ARFRP1 in carcinogenesis.
