Hexosaminidase B is the beta subunit of the lysosomal enzyme beta-hexosaminidase that, together with the cofactor GM2 activator protein, catalyzes the degradation of the ganglioside GM2, and other molecules containing terminal N-acetyl hexosamines. Beta-hexosaminidase is composed of two subunits, alpha and beta, which are encoded by separate genes. Both beta-hexosaminidase alpha and beta subunits are members of family 20 of glycosyl hydrolases. Mutations in the alpha or beta subunit genes lead to an accumulation of GM2 ganglioside in neurons and neurodegenerative disorders termed the GM2 gangliosidoses. Beta subunit gene mutations lead to Sandhoff disease (GM2-gangliosidosis type II). [provided by RefSeq, Jul 2008].

Making up nearly 6% of the human genome, chromosome 6 contains around 1,200 genes within 170 million base pairs of sequence. Deletion of a portion of the q arm of chromosome 6 is associated with early onset intestinal cancer suggesting the presence of a cancer susceptibility locus. Porphyria cutanea tarda is associated with chromosome 6 through the HFE gene which, when mutated, predisposes an individual to developing this porphyria. Notably, the PARK2 gene, which is associated with Parkinson's disease, and the genes encoding the major histocompatiblity complex proteins, which are key molecular components of the immune system and determine predisposition to rheumatic diseases, are also located on chromosome 6. Stickler syndrome, 21-hydroxylase deficiency and maple syrup urine disease are also associated with genes on chromosome 6. A bipolar disorder susceptibility locus has been identified on the q arm of chromosome 6. The C6orf64 gene product has been provisionally designated C6orf64 pending further characterization.

Mahogany (MG), originally identified as a protein involved in pigmentation, acts in conjunction with melanocortin receptors to suppress diet-induced obesity. Mahogany contains a single transmembrane domain, and it is expressed in a broad range of tissues, including the hypothalamus and pigment cells. Mutations within the mahogany gene were shown to rescue agouti-lethal-yellow mutant mice from obesity. The extracellular domain of mouse mahogany is the ortholog of the human protein attractin. Attractin (also designated DPPT-L) is a human serum glycoprotein and is a member of the CUB family of cell adhesion and guidance proteins. Attractin is expressed on activated T cells and is released from the cells 48 to 72 hours after activation.

A mitochondrial F1-ATPase inhibitor protein, ATPIF1 (ATPase inhibitory factor 1), also known as IP, IF1, ATPI or ATPIP (ATPase inhibitor protein), binds to the C-terminal region of a Beta subunit of the F1-ATPase at low pH values and, via interference of the Beta and Gamma subunit interaction, ATPIF1 regulates the activity of the F1F0-ATPase. This reversible ATPIF1 binding to F1F0-ATPase also occurs on the surface of endothelial cells.

CRISP-8 (Cysteine-rich secretory protein 8), also known as PI15 (Peptidase inhibitor 15), P25TI or SugarCrisp, is a 258 amino acid secreted protein that belongs to the CRISP family. Expressed at low levels in thyroid, prostate, salivary and mammary tissue, CRISP-8 functions as a serine protease inhibitor that exhibits weak inhibitory action against Trypsin, a serine protease found in the digestive system. In addition to its role as a protease inhibitor, CRISP-8 is secreted in neuroblastoma and glioblastoma cell lines, suggesting a role for CRISP-8 in tumor formation and metastasis within the central nervous system.

Defects in NOD2 are the cause of sarcoidosis early-onset (EOS) . EOS is a form of sarcoidosis manifesting in children younger than 4 years of age. Sarcoidosis is an idiopathic, systemic, inflammatory disease characterized by the formation of immune granulomas in involved organs. Granulomas predominantly invade the lungs and the lymphatic system, but also skin, liver, spleen, eyes and other organs may be involved. Early-onset sarcoidosis is quite rare and has a distinct triad of skin, joint and eye disorders, without apparent pulmonary involvement. Compared with an asymptomatic and sometimes naturally disappearing course of the disease in older children, early-onset sarcoidosis is progressive and in many cases causes severe complications, such as blindness, joint destruction and visceral involvement.
