TNF Alpha-IP 8L2 (tumor necrosis factor, alpha-induced protein 8-like 2), also known as TIPE2, is a 184 amino acid protein that shares 94% identity with its mouse counterpart and belongs to the TNFAIP8 family. Expressed in spleen, thymus, small intestine and lymph node with lower levels present in colon, lung and skin, TNF Alpha-IP 8L2 plays a role in maintaining immune homeostasis, specifically by acting as a negative regulator of both innate and adaptive immunity. In addition, TNF?IP 8L2 functions as a negative regulator of T-cell receptor function and is thought to promote Fas-induced apoptosis. The gene encoding TNF?IP 8L2 maps to human chromosome 1, which spans 260 million base pairs, contains over 3,000 genes and comprises nearly 8% of the human genome.

Acts as an adapter protein in a MDM2-DAXX-USP7 complex by regulating the RING-finger E3 ligase MDM2 ubiquitination activity. Under non-stress condition, in association with the deubiquitinating USP7, prevents MDM2 self-ubiquitination and enhances the intrinsic E3 ligase activity of MDM2 towards TP53, thereby promoting TP53 ubiquitination and subsequent proteasomal degradation. Upon DNA damage, its association with MDM2 and USP7 is disrupted, resulting in increased MDM2 autoubiquitination and consequently, MDM2 degradation, which leads to TP53 stabilization. Proposed to mediate activation of the JNK pathway and apoptosis via MAP3K5 in response to signaling from TNFRSF6 and TGFBR2. Interaction with HSPB1/HSP27 may prevent interaction with TNFRSF6 and MAP3K5 and block DAXX-mediated apoptosis. In contrast, in lymphoid cells JNC activation and TNFRSF6-mediated apoptosis may not involve DAXX. Seems to regulate transcription in PML/POD/ND10 nuclear bodies together with PML and may influence TNFRSF6-dependent apoptosis thereby. Down-regulates basal and activated transcription. Seems to act as a transcriptional corepressor and inhibits PAX3 and ETS1 through direct protein-protein interaction. Modulates PAX5 activity. Its transcription repressor activity is modulated by recruiting it to subnuclear compartments like the nucleolus or PML/POD/ND10 nuclear bodies through interactions with MCSR1 and PML, respectively.

CLLD6, is a 196 amino acid protein that contains one B30.2/SPRY domain. Expression of CLLD6 is found at highest levels in heart, skeletal muscle and testis, with lower levels found in thymus, peripheral blood leukocytes, lymph node and bone marrow. Existing as two alternatively spliced isoforms, CLLD6 maps to human chromosome 13, which comprises nearly 4% of human DNA and contains about 114 million base pairs and 400 genes. Key tumour suppressor genes on chromosome 13 include the breast cancer susceptibility gene, BRCA2, and the RB1 (retinoblastoma) gene. RB1 encodes a crucial tumour suppressor protein which, when defective, leads to malignant growth in the retina and has been implicated in a variety of other cancers. The gene SLITRK1, which is associated with Tourette syndrome, is on chromosome 13. As with most chromosomes, polysomy of part or all of chromosome 13 is deleterious to development and decreases the odds of survival.

NEEP21, also known as Brain neuron cytoplasmic protein 1, NSG1 (neuron-specific proteins family member 1), P21 or D4S234E, is a single pass type II membrane protein belonging to the NSG family. It is highly expressed during neuronal maturation but its expression is downregulated in adult tissues. NEEP21 predominantly localizes to Rab 4-positive early endosomes in the somatodendritic neuronal compartment and is essential for proper receptor sorting and recycling in neurons. It associates with GRIP1 and GluR-2 and mediates the surface expression of GluR-2. When this interaction is interrupted, GluR-2 accumulates in early endosomes and leads to changes in evoked synaptic current properties. In addition, NEEP21 forms a complex with the SNARE protein, Syntaxin 13 (also known as Syntaxin 12), and participates in the recycling of transferrin receptors (TFRs) and NTR2 (neurotensin receptor 2).
