The GK1.5 monoclonal antibody specifically binds with the mouse CD4 molecule, also known as L3T4, a 55 kDa differentiation antigen which binds to the MHC class II. CD4 is expressed on most thymocytes, a subpopulation of mature T lymphocytes, dendritic cells, pluripotent hematopoietic stem cells, B cell precursors, and lymphoid precursors inside the thymus. It is also expressed on the mouse egg cell membrane, enhancing adhesion to MHC class II bearing sperm. By interaction with MHC class II on the surface of APC, CD4 initiates the development of T lymphocytes and helps the optimum functioning of mature T lymphocytes. The binding of the GK1.5 antibody blocks the binding of the Anti-Mouse CD4 RM4-5 antibody.

The RPA-T4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation, together with the MHC class II. The RPA-T4 antibody is capable of blocking HIV binding and inhibiting syncytium formation, by binding to the D1 domain of the CD4 antigen. The OKT4 and the RPA-T4 monoclonal antibodies recognize different epitopes of CD4 and they do not exhibit cross-block binding.

KLRG1 is a receptor for cadherin, a family of transmembrane glycoproteins that mediate cell adhesion, and a common marker of T cell senescence. The receptor is believed to play an important role in the innate and adaptive immune system through the regulation of leukocytes. It is expressed on lymphokine-activated killer (LAK) cells, adherent LAK (A-LAK) cells, a sub-set of natural killer (NK) cells, T cells. In NK cells, it inhibits cytokine production and cytotoxicity activity.The receptor expression was not detected on the mouse peritoneal mast cells, or bone marrow mast cells.
