CD134 (OX40) is a T cell activation antigen structurally belonging to a lymphocyte-specific subgroup of the nerve growth factor and tumour necrosis factor receptor superfamily, which also includes the T cell antigen CD27, B cell antigen CD40, FAS antigen and the T cell activation antigen 4-1BB. The human CD134 protein is expressed only on activated CD4 + T blasts, and its ligand has been identified as gp34. Interactions between CD134 and its ligand in vivo are necessary for the differentiation of activated B cells into highly immunoglobulin-producing cells, however not involved in other pathways of antigen-driven differentiation of B cells such as development of memory cells in the germinal centers. In addition, the CD134 and gp34 system directly mediate adhesion of activated T cells to vascular endothelial cells, and contribute to growth stimulation of the virus-infected T cells.

PARP-1 (ARTD1) is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. It forms a complex with EEF1A1 and TXK that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production. PARP-1 (E998K mutant) is an inactive form of PARP-1 which can be used as a control compound.

CD40 is a Type I Transmembrane Glycoprotein that belongs to the TNF Receptor Superfamily. CD40 is expressed in B cells, follicular dendritic cells, dendritic cells, activated monocytes, macrophages, endothelial cells, vascular smooth muscle cells, and several tumor cell lines. The extracellular domain of CD40 is characterized by Cysteine rich repeat regions. Interaction of CD40 with its ligand (CD40L) leads to aggregation of CD40 molecules, which in turn interact with cytoplasmic components to initiate signaling pathways. Several different TRAF proteins (adaptor proteins) have been identified to serves as mediators of the signal transduction. CD40 plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
