Peptidyl-prolyl cis-trans isomerase NIMA-interacting 4(PIN4) is a peptidyl-prolyl cis/trans isomerase (PPIase) which interacts with NIMA and is vital for cell cycle regulation. PIN4 has 2 different isoforms: PAR14 and PAR17. Furthermore, PIN4 protein binds to double-stranded DNA under physiological salt conditions. PIN4 is involved as a ribosomal RNA processing factor in ribosome biogenesis. The PAR14 binds to tightly bent AT-rich stretches of double-stranded DNA, but PAR17 binds to double-stranded DNA.

B-Cell Linker Protein (BLNK) is a cell membrane protein which contains 1 SH2 domain. BLNK is expressed in B cells and fibroblast cell lines, playing a important role in B cell receptor signaling. BLNK as a central linker protein, downstream of the B-cell receptor (BCR), bridges the SYK kinase to a multitude of signaling pathways and regulating biological outcomes of B-cell function and development. BLNK associates with the activation of ERK/EPHB2, MAP kinase p38 and JNK, modulates AP1, NF-kappa-B and NFAT activation. BLNK involves in BCR-mediated PLCG1 and PLCG2 activation and Ca2+ mobilization and is required for trafficking of the BCR to late endosomes. BLNK deficiency results in agammaglobulinemia type 4 and much more profound block in B-cell development.

The TIM (T cell/transmembrane, immunoglobulin and mucin) family plays a critical role in regulating immune responses, including allergy, asthma, transplant tolerance, autoimmunity and the response to viral infections. The unique structure of TIM immunoglobulin variable region domains allows highly specific recognition of phosphatidylserine (PtdSer), exposed on the surface of apoptotic cells. Tim-1 (KIM-1; HAVcr-1) is a 100 kDa, type I transmembrane glycoprotein member of the TIM family of immunoglobulin superfamily molecules. There are two cytoplasmic alternate splice forms of Tim-1. One is a long (359 aa) kidney form termed Tim-1b, and one is a short (334 aa) liver form termed Tim-1a. Tim-1, important for asthma and allergy, is preferentially expressed on T-helper 2 (Th2) cells and functions as a potent costimulatory molecule for T cell activation.

Pre-B cell colony enhancing factor (PBEF) was originally identified as a cytokine that potentiated the clonal expansion and differentiation of pre-B cells, but it is also acknowledged to be the ubiquitous intracellular enzyme nicotinamide phosphoribosyltranferase (NAMPT) and the adipokine “visfatin”. PBEF is constitutively expressed in the fetal membranes where its greatest expression is in the amnion. It has intracellular and extracellular forms. Most of the intracellular functions of PBEF are due to its role as a Nampt which can induce angiogenesis through upregulation of VEGF and VEGFR and secretion of MCP-1. Extracellular PBEF has been shown to increase inflammatory cytokines, such as TNF- alpha, IL-1 beta , IL-16, and TGF- beta 1. PBEF also increases the production of IL-6, TNF- alpha, and IL-1 beta in CD14+ monocyctes, macrophages, and dendritic cells, enhances the effectiveness of T cells.

UCHL1 is involved both in the processing of ubiquitin precursors and of ubiquitinated proteins. This enzyme is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. Also,UCHL1 can bind to free monoubiquitin and may prevent its degradation in lysosomes. The homodimer of UCHL1 may have ATP-independent ubiquitin ligase activity. Furthermore, the mutations of UCHL1 gene can result in parkinson disease 5.

Interleukin-12 (IL-12), also known as Natural Killer Cell Stimulatory Factor (NKSF) or Cytotoxic Lymphocyte Maturation Factor (CLMF), is a heterodimeric pleiotropic cytokine made up of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit. IL-12 is produced by macrophages and B lymphocytes and has been shown to have multiple effects on T cells and Natural Killer (NK) cells. Some of these IL-12 activities include the induction of IFN-gamma and TNF in resting and activated T and NK cells; the enhancement of cytotoxic activity of resting NK and T cells, the stimulation of resting T cell proliferation in the presence of a comitogen; and the enhancement of NK cell proliferation. Current evidence indicates that IL-12 is a key mediator of cellular immunity and induces the differentiation of Th1 cells from precursor T helper cells. Based on its activities, it has been suggested that IL-12 may have therapeutic potential as a vaccine adjuvant that promotes cellular immunity and as an antitumour and anti viral agent.
