GPR proteins are integral seven-pass membrane proteins with some conserved amino acid regions. GPR19, an orphan receptor, shows elevated expression during embyronic development of the nervous sytem as well as in specific regions of adult mouse brain, including the olfactory bulb, the hippocampus, hypothalamic nuclei and the cerebellum. Beschreibung:   Lipoxygenases are a family of enzymes which dioxygenate unsaturated fatty acids, thus initiating lipoperoxidation of membranes and synthesis of signaling molecules, as well as inducing structural and metabolic changes in the cell. The Lox enzymes in mammals include 12-LO and 15-LO, which are classified with respect to their positional specificity of the deoxygenation of their most common substrate, arachidonic acid. The metabolism of arachidonic acid leads to the generation of biologically active metabolites that have been implicated in cell growth and proliferation, as well as survival and apoptosis. 15-Lipoxygenase (15-LO) acts in physiological membrane remodeling and the pathogenesis of atherosclerosis, inflammation, and carcinogenesis. It is highly regulated and expressed in a tissue- and cell-type-specific fashion. IL-4 and IL-13 play important roles in transactivating the 15-LO gene. Overexpression of 15-LO type 1 in prostate cancer contributes to the cancer progression by regulating IGF-1R expression and activation.

Belonging to the facilitator superfamily, HIAT1 is a 490 amino acid multi-pass membrane protein that may function as a sugar transporter and is expressed in adult and embryonic brain. The HIAT1 gene was first observed while analyzing for active genes in neonatal mouse hippocampus. The gene encoding HIAT1 maps to human chromosome 1, the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Beschreibung:   Dyrk (for dual specificity tyrosine phosphorylation regulated kinase) is the homolog of the Drosophila mnb (minibrain) gene, which is required for neurogenesis. Dyrk is a dual-specificity tyrosine kinase and serine/threonine kinase, which is itself regulated by tyrosine phosphorylation. Several mammalian Dyrk related proteins have been identified and are thought to compose a family of dual specificity protein kinases. Dyrk family members, including Dyrk1A (originally Dyrk), Dyrk1B, Dyrk1C, Dyrk2, Dyrk3, Dyrk4A and Dyrk4B, are thought to be involved in diverse cellular functions. Two isoforms of human fetal brain Dyrk2 exist: a deduced 528-amino acid protein and a protein containing 73 additional amino acids at the amino terminus. Dyrk3 is strongly expressed in testis, only after the onset of spermatogenesis, and very weakly expressed in spleen and adrenal gland. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Beschreibung:   Stanniocalcin 1 (STC1) and stanniocalcin 2 (STC2) are mammalian peptide hormones that were previously considered to be present only in bony fish, where they are involved in calcium homeostasis. STC1 plays a role in calcium and phosphate homoeostasis and is phosphorylated in vitro by protein kinase C, and STC2 is phosphorylated in vitro by casein kinase II (CK2). A human fibrosarcoma cell line, HT1080, expresses both STC1 and STC2 as secreted phosphoproteins in vivo, with STC2 being phosphorylated by an ecto-CK2-like enzyme. STC1 and STC2 have opposite effects on calcium and phosphate homeostasis, namely anti-hypercalcemic and anti-hypocalcemic actions, respectively. STC1 and STC2 are detected in human adrenal tumors, such as pheochromocytoma, differentiated neuroblastoma aldosterone-producing adenoma, and in cultured adrenal tumor cells (rat pheochromocytoma PC-12 cells and human neuroblastoma NB-1 cells).
