Chk2 Antibody: The p53 tumor-suppressor gene integrates numerous signals that control cell life and death. Several novel molecules involved in p53 signaling, including Chk2, p53R2, p53AIP1, Noxa, PIDD, and PID/MTA2, were recently discovered. The checkpoint kinase Chk2 is the mammalian homologue of yeast Cds1/Rad53. In response to DNA damage, the checkpoint kinase ATM phosphorylates and activates Chk2, which in turn directly phosphorylates and activates p53. Chk2 serves as ATM downstream effector to mediate activation of p53. Chk2 also phosphorylates and activates BRCA1, the product of a tumor suppressor gene that is mutated in breast and ovarian cancer.

ARMER Antibody: Apoptosis is important for normal development and tissue homeostasis. It is mediated by various caspases and ultimately results in the activation of endogenous endonucleases that degrade cellular DNA. Although apoptosis induced by endoplasmic reticulum (ER) stress is thought to be mediated by caspase-12, other caspases such as caspase-9 are also thought to be activated following ER stress. Recently, ARMER, a novel integral ER-membrane protein was shown to protect cells from ER stress-induced apoptosis. Analysis of the caspase proteolytic cascade suggests that ARMER acts by inhibiting caspase-9 activity, although the mechanism for this remains unkown. It should be noted that ARMER is not related to the inhibitor of apoptosis proteins (IAP) family and does not contain any baculoviral IAP repeat (BIR) domains.

RNF8 Antibody: RNF8 was identified as a ubiquitin ligase (E3) containing a RING finger motif and a FHA domain. This protein has been shown to interact with several class II ubiquitin-conjugating enzymes including UBE2E1/UBCH6, UBE2E2, and UBE2E3. RNF8 assembles at DNA double-strand breaks (DSBs) via interactions though the FHA domain with the adaptor protein MDC1, resulting in an increase in DSB-associated H2A histone ubiquitinations mediated by the associated ubiquitin ligase RNF168 followed by the accumulation of 53BP1 and BRCA1 repair proteins. Together with RNF168, RNF8 plays an integral part of class switch recombination in B cells, allowing the production of several classes of antibodies, through the recruitment of 53BP1 and BRCA1 to the DSB sites.

Friend leukemia integration 1 is a member of the ETS family of DNA binding transcription factors that is involved in cellular proliferation and tumorigenesis. Members of the Ets gene family share a highly conserved carboxy-terminal domain containing a sequence related to the SV40 large T antigen nuclear localization signal sequence. Approximately 90% of Ewing's Sarcoma (EWS)/Primitive Neuroectodermal Tumors (PNET) have a specific translocation, t(11;22)(q24;q12), which results in fusion of EWS to Fli-1, and production of an EWS-Fli-1 fusion protein. Among normal tissues only endothelial cells and small lymphocytes express Fli-1. This protein is expressed in majority of vascular tumors including angiosarcomas, hemangioendotheliomas, hemangiomas, and Kaposi's Sarcomas. High sensitivity and specificity of Fli-1 equals to or exceeds that of the established vascular markers like CD31, CD34, and Factor VIII.
