The retinal pigment epithelium (RPE) is a monolayer simple epithelium in proximity to the outer surface of the retinal photoreceptor cells. Retinal pigment epithelium-specific protein (RPE65) is a 65 kDa protein belonging to the carotene dioxygenase family. This protein is important in 11-cis retinal production as well as in visual pigment regeneration. RPE65 is attached to the membrane by a lipid anchor when palmitoylated (membrane form) and soluble when unpalmitoylated. The soluble form of the protein binds vitamin A. Defects in RPE65 causes autosomal dominant retinitis pigmentosa and/or Leber congenital amaurosis type 2.

HSP56 is a cis-trans prolyl isomerase belonging to the immunophilin protein family. The human HSP 56 gene (FKBP4) has multiple polyadenylation sites and the HSP 56 protein can undergo phosphorylation. HSP 56 influences immunoregulatory gene expression in lymphocytes, protein folding and trafficking. It can serve as a co-chaperone for steroid hormone nuclear receptors to govern appropriate hormone action in target tissues. The protein can associate with phytanoyl-CoA alpha-hydroxylase (PHYH) and with HSP90 through a series of tetratricopeptide repeat (TPR) domains. HSP 56 is a TRPC ion channel accessory protein that modulates channel activation following receptor stimulation.

A family of membrane bound, ubiquitous proteins involved in the selective transport of newly synthesized glycoproteins from the ER to the ERGIC include VIP36, ERGIC-53, ERGIC-1, ERGIC-2 and ERGIC-3. ERGIC-1, also designated ERGIC32, is thought to modulate the activity of a complex formed by ERGIC-2, also designated Erv41, and ERGIC-3, also designated Erv46. ERGIC-2 and ERGIC-3 are both mammalian homologs of yeast proteins abundant in COPII-coated vesicles and localize to the Cis-face of the Golgi apparatus.
