Nolz 1 is a 646 amino acid nuclear protein that is thought to function as a transcriptional repressor and is highly expressed in developing striatum. Additionally, Nolz-1 has been suggested to play a role in neural differentiation. A member of the Elbow/Noc family, Nolz-1 exists as three alternatively spliced isoforms and contains one C2H2-type zinc finger. The gene encoding Nolz-1 maps to human chromosome 10, which makes up approximately 4.5% of total DNA in cells and encodes nearly 1,200 genes. Several protein-coding genes, including those that encode for chemokines, cadherins, excision repair proteins, early growth response factors (Egrs) and fibroblast growth receptors (FGFRs), are located on chromosome 10. Defects in some of the genes that map to chromosome 10 are associated with Charcot-Marie Tooth disease, Jackson-Weiss syndrome, Usher syndrome, nonsyndromatic deafness, Wolman’s syndrome, Cowden syndrome, multiple endocrine neoplasia type 2 and porphyria.

KCNF1 is a multi-pass membrane-bound protein that acts as an ion channel and is generally expressed as a heterotetramer of potassium channeling proteins. Formerly known as kH1, KCNF1 is usually found as a heteromer with three other potassium channel proteins, KCNG3, KV6.3 and KCNV2. As a potassium channel protein, KCNF1 plays a role in regulating apoptosis and proliferation of pulmonary artery smooth muscle (PASM) cells. Bone morphogenetic proteins (BMPs) restrict proliferation and can induce apoptosis in normal human PASM cells and will upregulate expression of KCNF1 in PASM cells in vitro. KCNF1 is expressed in heart, brain, liver, skeletal muscle, kidney and pancreas.

Emp is a 396 amino acid ubiquitously expressed adhesion protein. Expressed as 5 alternatively spliced isoforms, Emp contains one CTLH domain and one LisH domain. Emp can form a complex with F-actin, which is involved regulating actin distribution in erythroblasts and macrophages. Considered to assist with cell division and nuclear architecture, Emp is localized with condensed chromatin at prophase, nuclear spindle poles at metaphase and in the contractile ring during telophase and cytokinesis. Although the exact function of Emp is unknown, Emp is suggested to be involvement in erythroblast-macrophage cell attachment, terminal maturation and enucleation of erythroid cells, and inhibiting apoptosis of erythroblasts.

The ubiquitin (Ub) pathway involves three sequential enzymatic steps that facilitate the conjugation of Ub and Ub-like molecules to specific protein substrates. The first step requires the ATP-dependent activation of the Ub C-terminus and the assembly of multi-Ub chains by the Ub-activating enzyme known as the E1 component. UBE2A (Ubiquitin-conjugating enzyme E2 A) and UBE2B (Ubiquitin-conjugating enzyme E2 B) are both Ub-conjugating enzymes that are essential to postreplication repair of UV-damaged DNA. UBE2A and UBE2B are both nuclear and cell membrane proteins that have been found to interact with Rad18.

Existing as multiple alternatively spliced isoforms that are expressed in neutrophils, eosinophils, monocytes and dendritic cells, CLEC-12A functions as a cell surface receptor that acts as a negative regulator of granulocyte and monocyte function and, via this activity, modulates signaling cascades. CLEC-12A is highly subject to post-translational glycosylation at its N-terminus and may also exist as a homodimer.
