BAF53b is a member of a family of actin-related proteins (ARPs), which shares significant sequence identity with conventional actins. Both actins and ARPs comprise an actin fold, which forms an ATP binding cleft. BAF53b is a component of the BAF (BRG/brm-associated factor) complex in mammals, which is functionally related to the SWI/SNF complex in S. cerevisiae and Drosophila, and is associated with chromatin remodelling to facilitate transcriptional activation. As neural progenitors exit the cell cycle, the highly homologous BAF53a subunit of the BAF complex is replaced by BAF53b. This subunit switch is essential for the transition from from neural stem cells / progenitors to postmitotic neurons. BAF53b also plays a role in dendritic outgrowth, during which it is required to recruit the BAF complex and the dendritic regulator CREST to specific gene promoters. This function can not be performed by BAF53a.

Fibroblast growth factors (FGFs) produce mitogenic and angiogenic effects in target cells by signaling through the cellular surface tyrosine kinase receptors. There are four members of the FGF receptor family: FGFR-1 (flg), FGFR-2 (bek, KGFR), FGFR-3 and FGFR-4. Each receptor contains an extracellular ligand binding domain, a transmembrane region and a cytoplasmic kinase domain (1). Following ligand binding and dimerization, the receptors are phosphorylated at specific tyrosine residues (2). Seven tyrosine residues in the cytoplasmic tail of FGFR-1 can be phosphorylated: Tyr463, Tyr583, Tyr585, Tyr653, Tyr654, Tyr730 and Tyr766. Tyrosine 653 and 654 are important for catalytic activity of the activated FGFR and are essential for signaling (3). The other phosphorylated tyrosine residues may provide docking sites for downstream signaling components such as Crk and PLCgamma.

Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain containing receptors, TNFR1 and Fas. Cell death signals are transduced by death domain (DD) containing adapter molecules and members of the ICE/CED3 protease family. A novel DD containing molecule was recently cloned from mouse, human and monkey and designated Daxx. Daxx is a death domain containing important intermediate in the Fas mediated apoptosis. Daxx binds specifically to the Fas death domain and enhances Fas induced apoptosis and activates the Jun N terminal kinase (JNK) pathway. It is widely expressed in fetal and adult human and mouse tissue, indicating its important function in Fas signaling pathways.

Parkinson's Disease, the second most common neurodegenerative disease after Alzheimer's Disease, is characterized by the loss of dopaminergic neurons and the presence of Lewy bodies (comprised of alpha synuclein and parkin inclusions). Autosomal Recessive Juvenile Parkinsonism (AR-JP) is a recently described form of Parkinson's Disease that has been linked to a gene that codes for parkin. Parkin, a 52 kDa protein, has a suggested role in the ubiquitin/proteasome pathway for protein degradation. The amino terminus bears sequence homology to ubiquitin while functionally it acts as a RING type ubiquitin protein ligase (E3) that coordinates the transfer of ubiquitin to substrate proteins, thus targeting them for degradation by the proteasome.

Epstein Barr virus (EBV) is a member of the herpesvirus family and one of the most common human viruses. Most people become infected with EBV during their lives. Primary infections usually results in infectious mononucleosis (glandular fever) but the virus can also lay dormant in B lymphocytes and when reactivated become associated with more serious disease such as Burkitt's lymphoma, nasopharyngeal carcinoma and Hodgkin's disease. EBV latently infects B lymphocytes. Infected B cells express EBV nuclear antigens and latent proteins LMP1, LMP2A and LMP2B. LMP2A forms aggregates in the plasma membranes of B lymphocytes, where it functions as a negative regulator of the Src and Syk protein tyrosine kinases. Studies show that LMP2A blocks B-cell receptor (BCR) signal transduction in EBV immortalized B cells in vitro and may play an important role in maintaining a latent EBV infection within the peripheral blood B cells of infected individuals.

The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. TANC (tetratricopeptide repeat, ankyrin repeat and coiled-coil domain-containing protein), also known as TANC1, is a 1,861 amino acid postsynaptic cell membrane protein that contains eleven ANK repeats, three TPR repeats and belongs to the TANC family. Considered a scaffolding component in the postsynaptic density, TANC interacts with TNIK, SAPAP1, Alpha-internexin, CaMKII, NMDA 2 and GluR-1. It is also thought that TANC interacts directly with SAP 97, PSD-95 and Homer. Upon stimulation by Rap 2, MINK1 and TNIK may phosphorylate TANC. The TANC gene encodes two alternatively spliced isoforms, contains approximately 264,025 bases and maps to human chromosome 2q24.2. Making up approximately 8% of the human genome, chromosome 2 consists of 237 million bases and encodes over 1,400 genes. A number of genetic diseases are linked to genes on chromosome 2 including Harlequin icthyosis, sitosterolemia and Alstré°‰ syndrome.
