The Interferon- alpha/ beta Receptor 1 (IFN- alpha/ beta R1) is a receptor which binds Type I Interferons including Interferon- alpha and - beta . It is a cell surface receptor and heteromeric receptor composed of one chain with two subunits referred to as IFNAR1 and IFNAR2. IFN- alpha/ beta R1, in association with IFN- alpha/ beta R2, is required for propagating antiviral signal transduction triggered by IFN- alpha and IFN- beta . IFN- alpha/ beta R1 interacts very weakly or not at all with type 1 interferons and does not stably interact with IFN- alpha/ beta R2. Ligands associate with IFN- alpha/ beta R2, and this complex subsequently forms a stable ternary assembly with IFN- alpha/ beta R1. IFN- alpha/ beta R1 also associates with IFN- gamma R2 even in the absence of IFN- gamma stimulation. Human IFN- alpha/ beta R1 contains a nuclear localization signal in its extracellular domain that is required for receptor translocation to the nucleus following interaction with ligand. Interferon stimulation results in an immunologic response that is especially associated with viruses.

Its function in the central nervous system is of interest, as mutations in the PGRN gene were found in cases of frontotemporal degeneration (FTLD). In addition, PGRN has also been linked to tumorigenesis. Progranulin is a biomarker for FTLD, other types of Alzheimer‘s Disease (AD) and potentially for MCI (Mild Cognitive Impairment). Additionally, PGRN is described as a new ligand of TNF receptors and a potential therapeutic against inflammatory disease like arthritis.

IRF6 is a member of the interferon regulatory transcription factor (IRF) family. Family members share a highly-conserved N-terminal helix-turn-helix DNA-binding domain and a less conserved C-terminal protein-binding domain. This protein is involved in palate formation.The protein encoded by this gene shares strong similarity with Saccharomyces cerevisiae Cdc23, a protein essential for cell cycle progression through the G2/M transition. This protein is a component of anaphase-promoting complex (APC), which is composed of eight protein subunits and highly conserved in eucaryotic cells. APC catalyzes the formation of cyclin B-ubiquitin conjugate that is responsible for the ubiquitin-mediated proteolysis of B-type cyclins. This protein and 3 other members of the APC complex contain the TPR (tetratricopeptide repeat), a protein domain important for protein-protein interaction.

The protein encoded by TIAL1 is a member of a family of RNA-binding proteins and possesses nucleolytic activity against cytotoxic lymphocyte target cells. The gene product is a cytotoxic granule-associated protein and has been shown to bind specifically to poly (A) homopolymers and to fragment DNA in permeabilized target cells. It has been suggested that members of this protein family may be involved in the induction of apoptosis. One isoform contains a lysosome-targeting motif in its C-terminal auxiliary domain; however, alternative splicing results in a T-cluster DNA-binding isoform, differing at the C-terminus where a hydrophobic sequence replaces the lysosome-targeting motif.

The recently identified protein suppressor of glucose by autophagy protein 1 (SOGA1) has been found to be involved in the regulation of autophagy. SOGA1 is thought to contribute to adiponectin-mediated insulin-dependent inhibition of autophagy during the activation of adenosine monophosphate kinase (AMPK) (1,2). SOGA1 contains an internal signal peptide that enables the secretion of a circulating fragment of SOGA1, providing a surrogate marker for intracellular SOGA1 levels. Knockdown of SOGA1 elevated glucose production in primary hepatocytes indicates that SOGA1 is an inhibitor of glucose production. It thus might be useful as a novel therapeutic target for diabetes.
