NCAM (neural cell adhesion molecule, CD56) is an adhesion glycoprotein with five extracellular immunoglobulin-like domains followed by two fibronectin type III repeats. Structural diversity is introduced by alternative splicing resulting in different cytoplasmic domains. NCAM mediates neuronal attachment, neurite extension and cell-cell interactions through homo and heterophilic interactions. PSA (polysialic acid) post-translationally modifies NCAM and increases the metastatic potential of small cell lung carcinoma, Wilms+ tumor, neuroblastoma and rhabdomyosarcoma. CD56 and CD16 are commonly used to identify NK cells although some cells with the T cell markers CD3 and CD4 also express CD56.

The 30-H12 monoclonal antibody specifically binds to MouseCD90.2, an alloantigen known as Thy-1.2, expressed on thymocytes, mature T cells, epithelial cells, neurons, hematopoietic stem cells, and fibroblasts. CD90 is a membrane glycoprotein that regulates the adhesion and signal transduction in T lymphocytes, and the adhesion of thymocytes to thymic stroma. The interaction between 30-H12 and the antibody to the CD3/TCR complex upregulates thymocytes signal transduction and apoptosis and downregulates mature T cell proliferation. The 30-H12 antibody seems to be unable to cross-link with CD90.1.

The GL-1 monoclonal antibody reacts specifically with the mouse B7-2 (also known as CD86), a costimulatory molecule expressed by B and T lymphocytes, macrophages (thioglycollate-induced from peritoneum), astrocytes and dendritic cells. The memory CD4+ T lymphocytes express CD86 (as mRNA and protein). CD86, and the B7-1 (CD 80) molecule, are ligands for CD152 and CD28, and influence the costimulatory interactions between lymphocytes B and T. B7-2 is also involved in the mouse natural killer cell-mediated cytotoxicity. The GL-1 antibody blocks the mixed lymphocyte reaction (MLR) and inhibits the T-cells stimulation by antigen-presenting cells. Mixtures of anti-B7-1 antibody and GL-1 were reported to inhibit the interaction between CD125 and its ligand with the in vivo priming of cytotoxic T lymphocytes.
